Solanum Lyratum Thunb. (Solanaceae), a herb of Solanum, which is one of the most valued Chinese traditional medicine, has been used as a herbal remedy for cancers, tumours and herpes over twenty centuries.Many researchers have studied its pharmacology, chemical constituents and clinical applications about Solanum Lyratum Thunb in the world. However, the study is concentrated in the saparation and structure identification, in which the analytical methods of the chemical constituents for Solanum Lyratum Thunb are scarce. The contents of herbal chemical constituents are important qualifications for their quality control and evaluation. Their analytical methods are important aspects of the modernization in traditional Chinese medicine. To exploit and utilize Solanum Lyratum Thunb effectively and reveal its pharmacological activity, our study is focused on the analytical methods of its chemical constituents which are simple, faster and accurate. The works involved are as follows:1 To develop an analytical method for polysaccharide. Solanum Lyratum Thunb was extracted with water after degreased by petroleum ether and ethanol. The absorbability of the extract colorated with phenol-sulfuric acid is in the point of 410nm by spectrophotometric method. Then, its polysaccharide content was calculated. There is a good linear relationship within the concentration range of 0.005~0.017mg·mL-1 ( r=0.9996). The average recovery of polysaccharide is 95.6% .The method is simple, sensitive and reproducible.2 To develop a GC method for Organic acid. Solanum Lyratum Thunb was extracted with chloroform by heating reflux. Then, samples were saponified, esterified, extracted with n-hexane and determined by GC. The chromatographic analysis was performed using HP-5 capillary column with FID detector. Highly pure N2 is used as carrier gas. Methyl Linoleate was separated well with other components. There is a good linear relationship within the concentration range of 0.02~0.14mg·mL-1 ( r=0.9997). The average recovery of Linoleic acid at high , medium and low concentration was 96.5%, 96.4% and 95.6% . 3 To develop the analytical methods for total flavonoids saponin. Solanum Lyratum Thunb was extracted with ethanol after degreased by petroleum ether. The content of total flavonoids in extract was ascertained by spectrophotometnc and HPLC methods respectively. The different results of the two methods were explained. Compared with HPLC method, spectrophotometric method is sensitive, good reproducibility and simpler, it's suitable to plant total flavonoids.4 To develop an analytical method for total saponin. Solanum Lyratum Thunb was extracted with methanol after degreased by chloroform. The absorbability of the extracts colorated with perchloric acid after extracted by n-butanol is in the point of 407.5 nm by spectrophotometric method. Then, its total saponin content is calculated. There was a good linear relationship within the concentration range of 0.002~0.012mgmL-1 ( r=0.9996). The average recovery of total saponin was 95.64%. The method is sensitive, good reproducibility, and suitable to routine analysis.
