Objective: To study the effect of lentina (LNT) on the expression of P53 protein, Bcl-2 protein and apoptosis in Lewis lung cancer-bearing Balb/C mice.Method : 40 Balb/C mice were devided into 4 groups randomly(n=10 each), Lewis lung cancer cell were implanted into all groups by subcutaneous injection. After 48h, the 4 groups mice were given 0.9%NaCl 0.2mL, LNT 5mg/kg, cisplatin 0.2mL(20mg/mL) and LNT 5mg/kg+cisplatin 0.2mL(20mg/mL) respectively, once a day. After the drug was given continuously for 10 days, the mice were killed by decapitation. Calculated the rate of inhibitory effect, observed the expression of P53,Bcl-2 protein by SABC immunohistochemistry method and apoptosis by electron microscope.Result: 1. General state of the 4 group: the mice of DDP had tarnished fur, small amout of food, spirit sagged, moved targily; LNT had more smooth fur and better psychosis, moved less targily than DDP; LNT+DDP had smooth fur, good psychosis, more food, moved agiliy. 2.Each group with drug had effect on inhibiting Lewis lung cancer tumor, LNT+DDP had the best effect, 46. 2%, the rate of inhibitory effect of LNT,DDP was 35.6%,39.7% respectively. 3.Under optical microscope, the cell of NS arrayed as nest, the nuclear was big, deep-stained, thickened-karyotheca, chromatin granulated, Pathologic karyokinesis can be seen. Proliferation of fibrous tissue, necrosis and infiltration can been seen in DDP group; There were little apoptosis , karyopycnosis,condensed-plasm, deep-stained and no infiltration in group LNT; More apoptosis in LNT+DDP; Under electron microscope the cell in NS was round or irregular, there were a lot of micro-villus on the cell surface, nuclear membrane was irregular with angular bites, many nucleolus, no swollen mitochondrion; the cell became smaller, mitochondrion was swollen, the chromatin crescent shaped along the inner margins of the nuclear; There was chromatinorrhexis in group DDP; There was karyopycnosis, chromatin condensation along the inner margins of the nuclear, more lysosomes could be found in cytosol, the mitochondrion became bigger and rounder, the mitochondrial crista became less and shorter, even disappeared. In condition of very swollen, it could became the structure of vacuole, and apoptosis body , which is the characteristic structure of apoptosis, could be found. 4. Anti-oncogene P53 protein was stained in nucleus with brown color, oncogene Bcl-2 protein was stained in cytoplasm, P53, Bcl-2 protein expression in LNT,DDP,LNT+DDP reduced after given drug.Conclusion: 1. LNT can raise the life quality of Lewis lung cancer mice. 2. LNT can inhibit the tumor of Lewis lung cancer mice. 3. LNT can induce apoptosis of Lewis lung cancer. 4. LNT can regulate the apoptosis-related gene P53> Bcl-2 expression. 5. Through inducing apoptosis, regulate the P53, Bcl-2 expression, LNT raise the life quartily and inhibit the tumor growth. 6. LNT have co-operation with cisplatin.
