Experimental Study Of Stasis To Stop Bleeding Side Effects Of Rat Mesangial Cell Proliferation

Objective: This article is composed to observe the effects that different dosages of medicated sera of the Zhuyu-Zhixue Prescription act on the proliferation, apoptosis and periodicity of the mesangial cells in vitro culture. It is also written to probe into the action mechanism and superiority of using this prescription in treating renal glomerular diseases.Methods: we use medicated sera prepared from experimental SD rats to acts on adherence-growing mesangial cells of rats and deviding the cells into 5 groups randomly which are called Normal Group, Tripterygium Glucosides Group, Zhuyu-Zhixue Prescription Group (High Dosage). Zhuyu-Zhixue Prescription Group (Mid Dosage) and Zhuyu-Zhixue Prescription Group (Low Dosage). And there is another group of mesangial cells without treatment of medicated sera called Blank Group in addition. Then we mensurate the numerical values of OD of the cells group by group in the 48 hour and the 72 hour by MMT afterward and calculate the rate of restrainability according to formularies. On the side we mensurate the rate of apoptosis and the DNA content of cellular periodicity by the flow cytometry. Results: 1.Every group of Zhuyu-Zhixue Prescription shows the effect of restrainability in proliferation of mesangial cells in the 48 hour and the 72 hour.2. Every group of Zhuyu-Zhixue Prescription shows the effect of promotion in apoptosis of mesangial cells in the 48 hour and the 72 hour.3. Every group of Zhuyu-Zhixue Prescription can restrain the caryocinesis of mesangial cells in different degrees in the 48 hour and the 72 hour and prevent the mesangial cells from proliferate in the G₁ period.Conclusion: It is proved that the Zhuyu-Zhixue Prescription have the effect onrestraining the proliferation of mesangial cells and provides credible experimentalwarrant on the clinical application of this prescription in treating Renal glomerulardiseases...
