The pathogenesis of ischemic stroke, as a clinical and critical disease, is complicated and also has a dangerous evolvement. This kind of disease causes serious threaten to the sufferers. Moreover, the sequelae, such as limbs dyskinesia, language barrier and decline of cognitive ability etc, bring heavy burden not only to the sufferers themselves, their families, and also the society. In our country, along with the development of economy, the improvement of living standard and the appearance of ageing problem, the incidence of that disease increase year after year. Thereby clinical and scientific researchers need to deeply study the pathogenesis of stroke and effectual therapy. The method of acupuncture cure stroke has a thousands history. Classical literatures as"Neijing"and"Zhenjiujiayijing"have correlative recordation. Especially in the period of convalescence, acupuncture is capable of accelerate the resume of organism function, like limb function, swallowing function and so on. And this method is also used clinically today.Empirical study confirm that the effect of acupuncture is versatility and multipath .it could improve the defect of animal neurosis caused by cerebral ischemia damage, could lower the degree of ischemia and anoxemia injury, could promote the injured region's recovery and functional restoration and so on. The recovery of blood circulation of ischemia semidarkness region, improvement of partly hypoxia state and prevention of more serious of impaired are key point in the stroke treatment. This experiment is designed to observe the blood vessel regeneration of ischemia injured region, and observe the variance of partly PCNA and VEGF expression. Combine with limbs functional recovery, we validate the therapeutical effect of acupuncture and investigate the possible mechanism of action.Objective: To observe the motor function variation of focal cerebral ischemical reperfusion injury rats'limbs at different time. To observe the expression of PCNA and VEGF. To observe the effect to that index when electric acupuncture the"Quchi"and"Housanli". To observe the effect to the focal cerebral ischemical reperfusion injury rats, which are inject eNOS catastaltic, when electric acupuncture that two acupoints. Through observing of above all things , we study acupuncture's blood vessel regenerated effect to cerebral ischemia reperfusion injury region, approach the possibility of brain functional recovery angiogenesis which promoted by acupuncture, provide new consider and experiment accordings for deepening acupuncture therapy of the ischemia stroke.Methods: (1) plug thread method to blunting rat's arteria cerebri media(MCAO model). (2) electric acupuncture"Quchi"and"Housanli"as therapeutic tool. (3) test lims motor function to observe the recovery of brain function. (4) immunity histochemistry method to observe PCNA and VEGF expression variation. (5) Selective restrain eNOS, observe the motor function variation of lims, the variation of PCNA and VEGF.Results: (1) focal cerebral ischemical reperfusion injury rats'limbs motor function is variable: other group rat's muscle strength test and grid experimental result is obviously different from normal group; acupuncture could improve the lims motor function, acupuncture group surpass model group; acupuncture function is weaken after injecting eNOS.(2)After reperfusion , PCNA masc cells is visible in brain. The PCNA number of acupuncture group in brain injured region is more than model group's. (3) VEGF masc cells is visible in ischemical reperfusion brain injury region. The VEGF number of acupuncture group is more than model group's. (4) After injecting eNOS, the number of VEGF is reduce. (5) when the ischemical reperfusion injury of the model group and acupuncture group is for 14 days, PCNA and VEGF's expression is reduced compared to 7 days group.Conclusion: (1) cerebral ischemical reperfusion injury rats'limbs motor function is variable. Acupuncture could promote lims'functional recovery. (2) PCNA masc cells is visible in cerebral ischemical semidarkness region. There is cell regeneration phenomenon. Acupuncture could strengthen injury region's PCNA expression, could profit injury recovery and functional reconstruction. (3) In ischemia semidarkness region for the model group and acupuncture group, PCNA masc cells percentage of 14days group is lower than 7days group. Along with the recovery of injury, cell multiplication is weaken. (4) In cerebral ischemia semidarkness region, there is VEGF masc cells and regeneration phenomenon. Acupuncture could strengthen injury region's VEGF expression, could profit protection after injury and blood vessel regenerate. (5) In ischemia semidarkness region for the model group and acupuncture group, VEGF masc cells percentage of 14days group is lower than 7days group. Along with the recovery of semidarkness region, ischemia and anoxemia state is getting improved, and VEGF is reduce. (6) As there are PCNA and VEGF masc cells in brain injured region, we could conclude that, after brain ischemical reperfusion injury, there are blood vessel regeneration phenomenon. Acupuncture could promote blood vessel regeneration, recovery blood supply sufficiently and quickly, and promote the recovery of brain injury region. (7)The VEGF masc cells percentage of inhibitor group is lower than acupuncture group. It state that the effect of acupuncture promote VEGF is partly depend on the existing of eNOS.
