Ischemic cerebrovascular disease (ICVD) is a kind of clinical common disease, and also frequently-occurring disease. Its attack rate both goes up year by year and makes more youthful. Following with the progress of medical diagnosis and treatment technic, the achievement ratio of salvage raised obviously,but the number of invalid also increased endlessly,result in incubus to the society. For the past few years, traditional Chinese medicine treatment by means of confirmative therapeutic effect, witch western medicine treatment cannot displace, to show tremendous preponderance on ICVD prevention and cure.This article comprise with records review and empirical study. In the part of records review: By look up and unscrambling pertinent literature document of ICVD in recent 10 years, first of all, to summarize the study overview of the cause, pathogenesis, diagnosis and treatment of this disease; then, to sum up the latest study advancement of the several chief immunizing cytokines witch participate the pathological process of ICVD; in the last place, to summarize the traditional Chinese medicine prehension of the cause and the pathogenesis and the advancement of the clinical treatment and empirical study of this disease.In the part of empirical study: To observe the morphologic changes and inflammatory reaction of rat model of focal cerebral ischemia at 12h and 24h and 3d, the changes of the contents of cytokines--IFN-γ,IL-6 and IL-8 in the circulating blood, and the influence of application of acupuncture on acu-points'Baihui'penetrate through'Qubin'combine with feed Naoxuekang for the above said indexes. The purpose is to study the regulatory function of Acupuncture Combined with Chinese Herbal Medicine therapy upon immune disorders at different time points in rat model of focal cerebral ischemia, and the regulating mechanism of acupuncture combine with Herbs therapy, thereby deepening the understanding of function and mechanisms of TCM therapy in treatment of ischaemic stroke.Methods(1) Occlude middle cerebral artery of rat by suture method for creating the model of acute focal cerebral ischemia (MCAO); (2) Observe neuron ischaemic necrosis of brain tissue, inflammatory immune response changes, inflammatory cell adhesion and infiltration by hematoxylin-eosin staining(HE);(3) Adopt double antibody with filling ABC-ELISA immunoassay method to detect the content of IFN-γ,IL-6 and IL-8 in blood serum.Results(1)In the early stage of focal cerebral ischemia, degeneration and necrosis of neuron, accumulation, adhesion and infiltration of inflammatory cell occurred. The scope and extent of brain tissue necrosis expanded and deteriorated with progressing of disease and continuing of inflammatory reaction. Acupuncture Combined with Chinese Herbal Medicine is able to reduce the inflammatory cell adhesions and infiltrations, and alleviate inflammatory immunoreactions and neuron necrosis. (2) In circulating serum in 12 hourses in cerebral ischemia, the IL-6 expression level increases obviously high, at lack 24 hourses in blood attains the high peak, later on slow-moving descent. Acupuncture Combined with Chinese Herbal Medicine contributes to the releases of IL-6 in serum at different time points, and the promoting function especially significant at 24h after ischemia.(3) The content level of IL-8 in serum gradually descended with the extension of ischemia time, while the content level of IFN-γin serum gradually increased. Acupuncture Combined with Chinese Herbal Medicine can help to restrain the releases of IL-8 and IFN-γin peripheral blood serum, and its restraining function is significant at 12h after ischemia, and as the ischemia time extended, the restraining function is more and more prominent.ConclusionAcupuncture Combined with Chinese Herbal Medicine treatment may enhance functional activities of anti-inflammatory cytokines and reduce the expression of the inflammatory cell cytokines in a certain scope to inhibit inflammatory cell accumulation and activation as well as releases of cell toxic substances, thereby oppressing the delayed neuronal damages, ameliorating the microcirculation, and promoting brain blood supply, contributing to the recovery of neural function. Therefore we can extrapolate, to effectively harmony the networks of various cytokines to weaken the inflammatory immunoreaction induced by the cytokines, thereby alleviating the cerebral ischemia damages, which is one of the most important mechanisms (paths) of Acupuncture Combined with Chinese Herbal Medicine to treat ischemic cereb ral vascular diseases.
