The Experimental Research on Effective Acupuncture Prescription in FocalCerebra Ischemic Rats Cerebrovascular disease is a kind of common disease in clinic attacked by highermorbidity and unfavorable prognosis, It is seriously harm for the human health due toits first disability and lethality rate among three major causes of death. Therefore, it isthe one of the most important project of the clinical and experimental research to studyphathophysiologic mechanisms and find the effective therapy for acute ischemic stroke. Objective:To observe the influence of acupuncure at different therapeutic timewindows,different therapeutic acupuncure point and different method of acupuncurestimulate in the focal cerebral ischemic injury in rats. Methods: (1) Use photochemical method inducing one-side middle cerebral arteryocclusion in rats to make focal cerebral ischemia experimental model. The acupuncuregroup were divided into 1h, 3h, 6h and 12h different therapeutic time windows,"Renzhong-point"use eletro –acupuncuncure, "Well points"use pricking method, treat6days. (2) Use neuroethological index to compare the symptoms of pretreatment andpost-treatment and thereby work out the statistical estimation (3) Use morphologicmethod to observe the inflammatory reaction in ischemic brain tissue. (4)observecalmodulin(CaM) activity in the ischemic brain tissues (5) observe the expressions ofBDNF in ischernic penumbra (6) Use radioimmunoassay method to assay the levels ofthe IL-1β, IL-6, IL-8 and TNF-αin the blood serum, which are used to analyze thetime-effect of acupuncture method. Results: 1. after acute cerebral ischemia, the inflammatory cell adhered and infiltrated topenumbra of the ischemic cortex, and this phenomenon aggravated as the ischerniacontinued. After treated acupuncture, the necrosis areas reduced and the degree of theadhesiveness and infiltration of the inflammatory cells also decreased. 2. When cerebral ischemia occurred 6days, CaM activity of the ischemic cerebralregion elevated distinctly in comparison with the normal control group and thesham-operation group(p<0.05). When cerebral ischemia occurred 6days, BDNF expression level in the modelcontrol group is low. there was no evidence in comparison with normal control groupand the sham-operation group(p>0.05). Following administration of acupuncure at fourdifferent therapeutic time windows, BDNF expression level were obviouslyincreased(p<0.05); Among of The acupunture group, the group of needling"Well-points"at 3 hours after ischemia and the group of needling "Renzhong"at 6hours after ischemia acupuncture were obviously increased. but there was no evidencebetween the different therapeutic time windows in BDNF expression level and therewas no evidence between the different acupunture point, too. 4. The levels of the IL-6,IL-8,TNF-αin blood serum increased significantly,which can be decreased by acupuncture significantly(p<0.05, p<0.01) ; The group ofneedling "Well-points"at 3 hours after ischemia-acupuncture were decreased obviouslyin comparison with the others therapeutic time windows(p<0.05). there was nosignificant difference between the different acupunture point, but "Well-points"acupuncture group were better than "Renzhong"acupuncture group by considering theaverage value of cytokine.
