Four Natures is the basic characteristic of Chinese Herbs. As one of the most important TCM theories Four Natures can guide the application of Chinese drugs and explain the principle of Chinese medicine therapy. There is important significance for the development of TCM to study Four Natures and establish the estimate model of Four Natures. The exploratory research on the relation between Four Natures and some physiological and biochemical indexes is carryed out by the means of pharmacological experiment and data mining.The objectives of this research are to reveal the relationship between Four Natures and some physiological and biochemical indexes , and to establish the estimate model of Four Natures.After consulting document we acquired the following research thoughts: give orally some Chinese Herbs which are cold or heat to animals, detect the physiological and biochemical indexe changes and analyze the data by the means of data mining.The tested drugs include Fuzi, Huanglian, Ganjiang, Dahuang, Xianmao, Zhizi, Rougui, Huangbo, Wuzhuyu, Tiandong. The tested animals include ICR Mice, SD Rats. The indexes measured in the experiments include: (1) general condition monitor:observation on the state, body weight, food consumption, water consumption; (2) central nervous system:spontaneous activity; (3) endocrine system:NO of testicle tissue, testicle coefficient, adrenal coefficient; (4) digestive system:gastrointestinal movement; (5) urinary system:urine volume, urine routine test; (6) cardiovascular system:blood pressure, heart rate; (7) energy metabolism:oxygen consuming content;animal body temperature, hepatic tissues ATPase, hepatic tissues SDH; (8) blood:blood clotting time, blood routine examination; (9) liver function:ALT, AST, AKP, Tbil, TP, GLB,ALB, A/G; (10) nephritic function:UREA, CR, UA; (11)lipid metabolism:TG, CHOL, HDL, LDL, NEFA; (12) others:TChE in whole blood; serum CK, serum GLU, serum T-AOC; thymus coefficient, lung coefficient, hepar coefficient, splenic coefficient.Summarize the common principles of the physiological and biochemical index Changes caused by cold drugs, and summarize the common principles of the physiological and biochemical index Changes caused by heat drugs. Cold-heat natures have close correlation with body weight, body temperature, ATPase, blood clotting time, WBC, PLT, MX%, MCH, serum ALT, serum UREA, serum CR, TChE in whole blood. More specific, cold nature has correspondence relation with body weight decrease, body temperature decrease, ATPase decrease, blood clotting time prolonged, WBC increase, PLT increase, MX% decrease, serum ALT decrease, serum UREA increase, serum CR increase; heat nature has correspondence relation with body temperature increase, ATPase increase, blood clotting time shortened, WBC decrease , PLT decrease, MX% increase, MCH increase, serum ALT increase, TChE in whole blood decrease.The data of animal experiments are processed by Bayesian Belief Network analysis. Main results are as follows: 12 indexes closely related to cold-heat nature are extracted from others and in the order of connected degree with cold-heat nature they are HDL, Tbil, testicle coefficient, hepar coefficient, CHOL, TP, RDW, MX#, Ca2+Mg2+-ATPase, RBC, MCH, TG. Further more,the relationship between cold-heat nature and 12 indexes is expressed by Bayesian Belief Network Topology Graph and Decision Tree. The preliminary estimate model of cold-heat nature based on the physiological and biochemical indexes is established.The result of this paper indicate that the combination of systematic experiments and data mining technique provide a more efficient method for the research and exploration of Chinese Herbs Four Natures.
