Colloidal particulate carriers have attracted considerable attention as a method of delivering drugs. Liposomes, as the most commonly used colloidal vehicle, are composed of a phospholipid bilayer that may carry for both hydrophilic and hydrophobic drug. Because of the unique properties, liposomes can improve efficacy and reduce toxicity for cancer treatment.In the present study, liposomes, loaded with methylcantharidimide, were prepared by the pH gradient-injection method. The prescription and preparation of the liposomes were optimized by orthogonal design. By electronic microscopy, the liposomes, prepared by optimized prescription, were large unilamellar vesicles, homogeneous shape with the particle size at 100nm~300nm, and the envelope efficiency was about 30%.Validated analytical method of HPLC for determining the conent of methylcantharidimide in the bio-samples of mice following i.v. administration of liposome and injection, respectively, were developed and used for evaluating the tissue distribution. Compared with injection, liposomes was characterized by a slower clearance, and a remarkable increase drug in liver. These results indicate that methylcantharidimide Liposomes has liver targeting property.
