Electroporation is a new transdermal drug delivery method which was studied in the field of transdermal drug delivery system recently. But the studies about electroporation were all use single compounds, there were not studies which use traditional Chinese medicine as model drug.The objective of the experiment was to study influences of electroporation and penetration enhancers on the transdermal transport of Qingfengteng. And studies the preclinical pharmacokinetics of the model drug in animals, we draw the conclusions as follows:(1) Firstly we found that traditional Chinese medicine could transport across human skin to therapeutic levels use electroporation, however, the behaviors of the traditional Chinese medicine transport across human skin were different from that of single chemical compounds. Previous studies suggested that electroporation could happen if the voltage higher than 100V, but it could happen under 100V for traditional Chinese medicine.(2)Triangular wave pulses(TW), saw-toothed wave pulses(STW) and some allied wave pulses which were first reported in the world, we found the differences of wave form of pulses have caused significantly different fluxes of drug.(3) Bohe Volatile Oil was divided into B1, B2, B3, B4, B5 and B6 under the vacuum situation. Irritation of B1, B2, B3, B4, B5 was lower than that of Bohe Volatile Oil; while the irritation of B6 was higher than that of Bohe Volatile Oil; We also found the higher the vacuum situation temperature the severer the irritation. There was a considerable synergism between electroporation and B4 or B6.(4) Firstly we found that during electroporation some electrolyte ions enhanced the transdermal delivery of drug by electroporation, and the synergistic advantages was not affected by the position of the cathode and anode.(5) Through the studies of preclinical pharmacokinetics of drug in animal, as a result, the Cmax and AUC0â†'8 of electroporation was larger than that of passive diffusion; MRT and Tmax of electroporation were reduced compared with passive diffusion.Our work demonstrated the feasibility of transdermal delivery of traditional Chinese medicine by electroporation, if we choose the better electrical parameters and appropriate permeation enhancer would increased the efficiency of electroporation for transdermal drug delivery. Combing optimized electroporation parameters and better permeation enhancers may be a promising drug delivery system for traditional Chinese medicine.
