Folium Eucommiae are dry leaves of Eucommia ulmoides. Eucommia barks are used as medicine traditionally, but the barks are limited, eucommia will die without barks. To protect the eucommia resources, some people are studying on the chemical constituents of the leaves of Eucommia ulmoides, there are many achievements now. This profeeional subject is made of three parts. Part one is the review of Eucommia ulmoides leaves.The second part mainly introduce a reliable and exact method of HPLC to determine the content of chlorogenic acid. In addition to, we improve the extraction artwork of chlorogenic acid. In part three, we amended the method of HPLC to determine the contnents change of aucubin, chlorogenic acid and PDG, during heating.The Eucommia ulmoides leaves' pharmacologic action and chemical composition are similar to the barks', they all have depressurization. This subject applies some method, such as ultrasound, merceration, column chromatography and rotation lamellar to extract effective component. Four compounds were achieved using isolation methods: A1, A2, B, C. Two of them were elucidated by the means of spectroscopy (UV,IR,NMR,MS etc.), physical and chemical constant, reference substance. Compound B is aucubin, C is chlorogenic acid. Compound A1, A2's polarity is similar to each other, so we can't isolate them with column chromatography. We apply rotation lamellar to isolate them, and we obtain compound A1. Lastly, we use preparative lamellar to purify compound A2. The prity of compound A1, A2 are 99.5%, 99.2%. The frameworks of compound A1, A2 are identifying.Chlorogenic acid is one of the active components of Eucommia ulmoides leaves. It has many distinct pharmacologic actions, such as antibiosis, eliminate inflammation, anti-mutagenesis and anti-malignant tumor. HPLC is the main method at present. This method is simple and accurate. It is seldom affected by human factor, its usage is very generally. This text amended a reliable and exact method of HPLC to determine the content of chlorogenic acid. On the fundament of the study, we improve the extraction artwork of chlorogenic acid by lime cream precipitation method, boiling with water. We also made an intermediate test. The extraction percentage of chlorogenic acid is 44.7%. This proof that the method refers to industry produce.Furthermore, we study the contents change of aucubin, chlorogenic acid and pinoresinol diglucoside in barks and leaves of Eucommia ulmoides with HPLC, during heating. The contents of the aucubin and chlorogenic acid in the leaves which were in boil water for 5 minites and then dried in 60℃, were much higher than those in the traditional processing products. However, the content of the aucubin in the barks which were dried in 80℃was the highest in all, the contents of the chlorogenic acid and the PDG in the barks had no abosolutely change. This method can offer references for the processing and saving up Eucommia ulmoides.
