In recent years, there are increasing trendency of spinal cord injury. Because SCI can lead to a very high disablement rate—completely or partly function loss of aesthesis, action an urine and stool, now it is still a hot and difficult problem for scientists of every country that how to protect the injured cord effectively and promote its recovery. According to thestudies recently, Apoptosis is an important way of secondary cell death in SCI. Among of them, Caspase-3 is the final executant ofapoptosis. It is an important part of SCI treatment that how to interfere in it.Objective: To observe the inhibitory action of velvet antler polypeptide to induce fetus rat spinal nerve cell to apoptotic to radiation.Method: Flow cytometry detection the effect that the spinal nerve cell apoptotic that different velvet antler polypeptide dosages wither to the cell after 2Gy irradiate ;The flow cytometry and immuno-histochemistry detection the spinal nerve cell after 2Gy irradiate , the velvet antler polypeptide to apoptotic inhibitory action.Result :The fetus rat spinal nerve cell is unlike dosages by that 2Gy linear accelerators irradiate the velvet antler polypeptide that 0,50mg/ml,100mg/ml,200mg/ml, 400mg/ml, 800mg/ml apoptosis percentages is 41. 51%±13. 68,35.75%±11.90,26. 46%±10.30,24. 61%±10. 87,20. 10%±8. 54,20. 64%±8.96。Contrasting the group, the cell of irradiation (2Gy) group , irradiate (2Gy) + the velvet antler polypeptide (400 mg/ml) group and irradiate (2Gy) + NGF (20μg/ml) group apoptosis percentages is 14.12% + 5. 01 , 41.51% +13.68 , 20.10% +8.54 , 19.78% +6.60, irradiations (2Gy) + velvet antler polypeptides (400mg/ml) have the remarkable difference with simple irradiations (2Gy) group (p<0.05). There is no difference comparing with irradiations (2 Gy) + NGF5μl (20ng/μl) group. immuno-histochemical caspase-3,Bcl-2,Bax of the velvet antler polypeptide group. The masculine cell number reduction have remarkable difference with simple irradiations group (p<0. 05). There is no difference comparing with NGF group.Conclusion: The velvet antler polypeptide has the effect restraining radiation induce the spinal nerve cell to apoptosis.
