Objective:To investigate the relationship between microsatellite alteration of RASSF1A gene and infection of HPV16and the development of cervical carcinoma.Methods:We selected two sites of microsatellite polymorphism of RASSF1A gene(D3S1478 and D3S4604),used polymerase chain reaction(PCR)technique and perpendicular electrophoresis technique,to detect the LOH and MSI of 110 cases formalin-fixed paraffin-embedded samples(including 50 cases of cervical carcinoma,40 cases of cervical intraepithelial neoplasia and 20 cases of normal cervical tissue).Meanwhile we detected the infection state of HPV16in cervical samples.The datas were analyzed through SPSS13.0 software package,numeration data usingχ2 test and Fisher's exact propability.Results:(1)At D3S1478 and D3S4604,the LOH and MSI incidence rates of cervical carcinomas were 32.6%,14%and 48.9%,19.1%,respectively.The LOH and MSI incidence rates of CIN were 31.4%,11.4%and 39.5%,15.8%,respectively.There was no significant difference between cervical carcinoma and CIN refered to their incidence rates of LOH and MSI at the two sites.(2)The LOH of RASSF1A gene at D3S1478 and D3S4604 had obviously differences between stageⅠ~ⅡandⅢ~Ⅳof cervical carcinomas,moreover between the well/moderately differentiated cervical carcinomas and the poorly differentiated cervical carcinomas(P<0.05).Significant differences were also existed between the cervical carcinomas with lymph node metastasis and those without lymph node metastasis in regard to their LOH and MSI incidence rates at the two sites(P<0.05).The incidence rates of LOH and MSI in CINand cancer in situ were higher than that in CINⅠ/Ⅱ(P<0.05).(3)The positive rates of HPV16infection in cervical carcinomas were obviously higher than that in CIN and normal cervical tissues(P<0.05),and the incidence rates of LOH of RASSF1A gene were more higher in HPV16(+)than that in HPV16(-)(P<0.05).Infection of HPV16and LOH of RASSF1A gene may have reacted together in the development of cervical carcinoma.Conclusions:There were some relationships between the abnormal expression of RASSFIA gene and the increasion of HPV16infection in cervical carcinomas.The change of RASSF1A gene is a relatively late event in cervical carcinomas.The detection of the LOH and MSI of RASSF1A gene might be helpful to the early diagnosis and the screening of cervical carcinoma.It might also be useful for predicting the prognosis of cervical carcinoma.
