Patients With Psoriasis Keratinocyte Cells To T Cells, Cla And Cd45ro Expression In Rats

Objective:To reveal the action of psoriatic keratinocytes on T lymphocytes in CLA andCD45RO protein expression and the significance in the pathogenesis of psoriasis.Methods:Density gradient centrifugalization and absorption technique were respectively applied to detach peripheral blood mononuclear cells(PBMC),monocytes and peripheral blood lymphocytes(PBLC),T lymphocytes were got by Nylon post absorption method.Monocytes inactived by ⁶⁰Co-γirradiation were used APC.Keratinocytes inactived by 60Co-γirradiation were cocultivated respectively with T lymphocytes and monocytes for 72h.Flow cytomety(FCM)technique were performed to detect the expression of CLA and CD45RO proteins.Results:There were significant overexpression of CLA proteins in T lymphocytes cocultivated with psoriatic keratinocytes,were significantly different compared with the psoriatic group. Expression of CLA proteins in T lymphocytes cocultivated with keratinocytes were significantly different compared with the normal T lymphocytes group.The expression of CD45RO in T lymphocytes cocultivated with psoriatic keratinocytes were higher than others..Conclusion:Psoriatic keratinocytes can induce T lymphocytes to activation..The expression of CLA and CD45RO proteins might be one of the important mechanisms.Psoriatic keratinocytes have much more significant influence on CLAandCD45RO protein expression of T lymphocytes, which might be responsible for early and last psoriatic lesions formation.
