Introduction:Thyroid papillary carcinoma is frequent cancer in the head and neck tumor,and invasion and metastasis metastasis of tumor is the main reasion of poor prognosis.Recently,a structurally distinct family of membrane glycoproteins including MRP1/CD9,CD37,CD53,ME491/CD63 and KAI1/CD 82,has been called transmembrane 4 superfamily proteins(TM4SF).Although the precise biological function of TM4SF are still unknow,a substantial body of evidence has shown that TM4SF members are involved in regulating cell adhesion,motility and proliferation.Two members of the TM4SF,KAI1 /CD82(Kang-ai-1)and MRP1/CD9(motility-related protein)has been reported to be associated with the metastatic potential of some tumors,including prostate cancer,breast cancer and lung cancer.However,report of the expression of KAI1/CD82 and MRP1/CD9 gene in thyroid papillary carcinoma is the expression of KAI1/CD82 and MRP1/CD9 in thyroid papillary carcinoma is least understood.Objective:To investigate the relationship between expression of KAI1/CD82 and MR P1/CD9 and invasion and metastasis in thyroid papillary carcinoma.Methods:1.Amultiplex reverse transcriptase-polymerase chain reaction method were used to examine the mRNA expression of the KAI1/CD82 and MRP1/CD9 in 44 thyroid specimens,including 24 thyroid papillary carcinomas(PTC),2 undifferentiated carcinoma(UTC),10 follicular adenoma,8 multinodular goiters,and 8 normal thyroid tissue.Finally,whether expres sion levels of KAI1/CD82 and MRP1/CD9 are associated with specific clinicopathologic characteristics of PTC patients was determined. 2.Immunohistochemical methods(EnvisionTM)were used to examine the expression of KAI1/CD82 and MRP1/CD9 in paraffin-embedded specimens in 94 specimans,including 63 PTC,3 UTC,follicular carcinomas(FTC),5 thyroid carcinoma medullare(MTC),13 follicular adenoma,8 multinodular goiters,and 8 normal thyroid tissue.The results were compared.Results:1.The expression level of KAI1/CD82 mRNA was 3.59±1.57 and the positive rate of protein was 46.03%(29/63)in PTC,both of which were and protein in PTC was inversely related to the follicular adenoma (P<0.01,P<0.05),multinodular goiters(P<0.01,P<0.01),and the expression of KAI1/CD82 mRNA and protein were higher in PTC than in normal thyroid tissue(P<0.01,P<0.01).There was also a significant correlation between KAI1/CD82 mRNA and protein expression and the lymphaden metastatic (P<0.05)and the tumor size(P<0.05).In addition,the expression of KAI1/CD82 mRNA and protein were total coincident(r=0.486,P<0.01).2.The expression level of MRP1/CD9 mRNA was 1.41±0.47 and the positive rate of protein was 41.27%(26/63)in PTC,both of which were inversely related to the follicular adenoma(P<0.05),multinodular goiters(P<0.05),and normal thyroid tissue(P<0.05).There was also a significant correlation between MRP1/CD9 mRNA and protein expression and the invasion and lymphaden metastatic of tumor(P<0.05).In addition,the expression of MRP1/CD9 mRNA and protein were total coincident(r=0.465,P<0.01).3.There was also a significant correlation between KAI1/CD82 and MRP1/CD9 mRNA in 24 PTC(r=0.399,P<0.05).In addition,the co-expression of KAI1/CD82 and MRP1/CD9 protein were significant correlation with the lymphaden metastatic of tumor(P<0.05).Conclusions:1.The expression level of KAI1/CD82 mRNA and the positive rate of prot- in in PTC are significant higher than follicular adenoma,multinodular goiters and normal thyroid tissue,which are related with the carcinogenesis of in thyroid papillary.2.The expression level of MRP1/CD9 mRNA and the positive rate of protein are lower progressively of PTC than those of normal thyroid tissue, multinodular goiters and follicular adenoma,which are related with the carcinogenesis of in thyroid papillary.3.In this study,we find that the expression level of mRNA and the positive rate of protein are positive correlation between KAI1/CD82 and MRP1/CD9.The decreased expression of expression of KAI1/CD82 and MRP1/ CD9 are negative correlation with the invasion and lymphaden metastatic of tumor.Our results indicate that KAI1/CD82 and MRP1/CD9 has tumor metastasis depressant effect.KAI1/CD82 and MRP1/CD9 may play an important role in development and metastasis of PTC,and they may be used as potential markers for lymphaden metastasis potential of PTC.
