Qingwen Huayu Decoction Anti-influenza Virus Experimental Study

Objective: Making full use of the abundant medicine herbs in the tu and miao minority nationalities regions, to screen the effective clearing heat and antiviral prescription. In the base of first-phase researches, to compose compound prescription from which were reported have identifiable antiviral action-QingWen huayu liquid. Through biological experiment, to evaluate the anti-influenza: virus activity and immunological regulation effection of one effective prescription in mice ,and to observe it' s clearing heat effcetion in rat. with the purpose to cure the upper respiratory viral infection such as influenza, especially to supply theories for its clinical application and empirical study. Besides, the research report is beneficial to prevent and control of diseases such as sever acute respiratory syndrome, it's also beneficial to full develop and use original medicine herbs of the areas inhabited by the tu and miao minority nationalities.Methods: 1. Antivirus exponent in vivo: viral pneumonia model were replicated by dribbling nose with Influenza FM1 in the mice, Study of QingWen huayu liquid influence on the lung and spleen index number, pathology of the lung tissue and death rate of mice infected with influenza virus . 2. Study of QingWen huayu liquid inf luence on the body' s immunity of the mice infected with influenza virus: The expressions of cytokines Tumor necrosis factor-a(TNF-a),Interleukirr(IL-8) in the mice plasma assayed by radio-immunity. To observe QingWen huayu liquid influence on the phagocytizing function of mice' s abdominal macrophages using purge blood serum of carbon ink method. 3. Clearing effection: By establishing the rat of body high temperature witch caused with dried yeast animal model, observed the temperature' s changes in different phases.Results: In vivo, QingWen huayu liquid could improve the mice pneumonia caused by influenza virus (Comparison with model control group p＜0. 01), especially the high dose group (15g/kg. d) could depress the lungs index obviously, The effect of large dose group (15g/kg. d) is similar to virazole (p＞0.05), and more effective than antiviral liquor (p＜0.05); There was no obvious difference of spleen index among groups(p＞0. 05).Compared with control group, TNF-a and IL-8of the model group(p＜0.05). TNF-a and IL-8 of prophylaxis groups decreased greatly contrasted with model group(p＜0.05);Death protection of the herbal medicine on influenza FM1 infected mice: The result shows that the mortality rate of infectious model group reaches 75% and virazole group, 25%, significantly higher (p＜＜0.01)than model group, QingWen huayu liquid middle dose group' s mortality rate is 32% and the low dose group' s mortality rate is 37.5%, significanftly better than model group' s(p＜0.05). QingWen huayu liquid large dose group' s mortality rate is 50%, compared with model group, The effect of large dose group's is similar to model group's (p＞0.05). Study of QinggWen huayu liquid' s influence on the phagocytizing function of influenza FM1 infected mice' smacrophages: the result shows that phagocytizing function of infectious model group is lower than control group(p＜0.05) , the large and middle dose group is a bit higher than infectious model group(p＜0. 05, p＜0.01). Study of the herb medicine' s influence on the rat' s temperature: the result shows that QingWen huayu liquid can reduce the rat's temperature, especially the large and middle dose group had significant differences with the blank control group(p＜0. 05, p＜0. 01).Conclusions: QingWen huayu liquid can improve the symptom of mice pneumonia caused by influenza FM1 and improve the inflammation of the lung tissue, and also can inhibit the expressions of cytokines TNF-a and IL-8 in the serum of influenza FM1 infected mice. QingWen huayu liquid can reduce the mortality rate of influenza FM1 infected mice, prolonging their lives, and enhance the nonspecific immunitical function; also the clearing heat is mainly reason. Quibus, the results indicate that the QingWen fluayu liquid can educe the synergism for antiviral suppression. The experiment supplied the theory according for the clinical application and further empirical study in future.
