Objective:1. Observe the intestinal barrier function of SD rats treated with 5-Fu intraperitoneal injection , compare the effect of different scheme of 5-Fu administration on the intestinal barrier.2. Investigate the role of Bifidobacterium longum in the protection of the intestinal barrier function in such situation.Method:72 SD rats were divided into six groups, each group with 12, they were Control group of low-dose(A), 5-Fu in low dose (B), Bifidobacterium longum with low dose 5-Fu (C), Control group of high-dose(D), 5-Fu in high dose (E), Bifidobacterium longum with high dose 5-Fu (F). Low dose groups daily injection 5-Fu 40mg/kg intraperitoneal D1-5, high dose groups injection 5-Fu 500mg/kg intraperitoneal on the first day, control groups inject saline instead. Bifidobacterium longum were given to group C, group F orally D1-7,other groups were given saline instead. The weight and diarrhea were data everyday. On the fourth day and on the eighth day killed 6 rats in each group , to investigate the pathological changes of intestinal mucosa and the expression of Occludin, PCNA, sIgA.Results:1. WeightRats in group A and group D body weight increased daily while others loosed body weight continuously. On the fourth day the weight lose of rats in group B,C,E,F was significantly compared with control groups P<0.05, the weight loss of rats in group C was less than other three groups P<0.05. On the eighth day, weight of rats in group B,C,E,F dropped further, the weight loss of rats in group F less than other three groups P<0.05.2. DiarrheaOn the fourth day ,Rats in group B,C,E,F, suffered from diarrhea. The situation in rats of group B was worse than that in group C, group E was worse than that in group F, group E was worse than that in group B, The situation in group E was worst.P<0.01. On the eighth day ,Rats in group B,C,E suffered from diarrhea. The situation in rats of group B was worse than that in group C, group E was worse than that in group F, group E was worse than that in group B, The situation in group B was worst.P<0.01.3. Pathological scoreOn the fourth day , the pathological score in rats of group B was higher than that in group C, group E was higher than that in group F, group E was higher than that in group B, Group E got the highest score and the intestinal mucosa was damaged the most severely ,P<0.01. On the eighth day , the pathological score in rats of group B was higher than that in group C, group E was higher than that in group F, group B was higher than that in group E, Group B got the highest score and the intestinal mucosa was damaged the most severely ,P<0.01.4. The expression of occluding ,sIgA ,PCNAGroup B,C On the fourth day , the expression of occluding and sIgA decreased and the expression of PCNA increased , compared to the control groups P<0.05. In group C the expession of occluding and sIgA and PCNA was higher than that of group B ,P<0.05. On the eighth day, the expression of occluding and sIgA decreased further, the expression of PCNA went down, In group C, the expression of occluding ,sIgA and PCNA was higher than that in group B.Group E,F On the fourth day , the expression of occluding ,sIgA decreased and the expression of PCNA increased , compared to the control groups P<0.05. In group C the expession of occluding ,sIgA and PCNA was higher than that in group B ,P<0.05. On the eighth day, the expression of occluding ,sIgA increased, the expression of PCNA went down, In group F, the expression of occluding ,sIgA was higher than that in group B. and PCNA was lower than that in Group B.P<0.05.Group B,E Group C,F On the fourth day , the difference of level in expression of occluding ,sIgA ,PCNA between group B and E,between C and F was not significantly . On the fourth day , the difference of level in expression of occluding ,sIgA ,PCNA between group B and E,between C and F was significantly.Conclusions:1. Rats treated with the injection of 5-Fu intraperitoneal can successfully found the model of intestinal barrier dysfunction, the animals got diarrhea and loosed their body weight significantly. The intestinal mucosa necrosis and the expression of occludin ,sIgA in the intestinal tissue decreased. In the rats of given 5-Fu 500mg/kg once, the damage to de intestinal mucosa can be much recovered in one 7 days.The administration scheme of 5-Fu injection intraperitoneal as 40mg/kg for 5 days caused much severe damage to the intestinal mucosa than the scheme of 500mg/kg at once.2. Given Bifidobacterium longum orally can protect the intestinal mucosa in the rats suffered from 5-Fu injection. They were helpful to keep the body weight ,relieve diarrhea ,increase the expression of Occludin and sIgA in the intestinal gut, rapid the repair of the intestinal mucosa.
