The dissertation includes two segments:·To study chemical constituents in Potentilla discolor Bunge. and validate the activities of chemicals.Using information technology, virtual screening based on 3-D pharmacophores was applied to predict the activities of chemicals in Chinese materia medica. The chemicals were isolated from Chinese materia medica, guided by the results of virtual screening. Through muti-level pharmacodynamic experiments, the results calculated by computer, based on 3-D pharmacophores, were investigated.The result, calculated by vitual screening, based on 3-D pharmacophores for the activity on anti-inflammation, indicated that tormentic acid has the activity. Literatures show that tormentic acid exists in Potentilla discolor Bunge.. Therefore chemical constituents in Potentilla discolor Bunge. were study, six compounds were isolated by chromatography, and three of their structures were elucidated by spectral analysis. Tormentic acid was isolated and identified. Content of total triterpenoids was also determined by ultraviolet spectrophotometry. Tormentic acid was applied to the screening of in vitro activity on anti-inflammation.Through detecting LTBB4, metabolites of AA, it concluded that tormentic acid had no inhibition on activity of 5-LOX. Arachidonic acid and its metabolites play an important role in pathogenesis of inflammation. Leukotrienes is a kind of metabolites of arachidonic acid, hydrolyzed by 5-lipoxygenase, discovered in the late 1970s. It's highly inflammatory mediators, participating in pathogenesis of gout, asthma and rheumatoid arthritis and so on. Searching for inhibitor on leukotrienes is an important means to develop a new anti-inflammatory drug. We applied tormentic acid to the screening of in vitro activity on anti-inflammation, no inhibitory action on metabolic pathway of AA was observed.·Samples containing water quantified by NIR Spectroscopy technique NIR spectroscopy technique was applied to quantify glucose in samples containing water to investigate the feasibility of quantifying samples containing water. The indices of RMSEV,RMSEP,r2 was applied to evaluate models with different preprocessed spectrum in different regions. The wavelength range of 9931.60cm-1 and 7266.46cm-1 was selected to develop calibration model with first derivative spectrum. Model calibrated indicates a RMSEP of 0.075 and r2 of 0.99925 for glucose without intervention of strong absorption of water, is suitable for quantifying glucose in samples containing water. Therefore this technology is nondestructive,fast,without taking of sample, can be taken as a potential method for quantifying samples containing water.
