Pulsatilla chinensis(Bge)Regel is a Pulsatilla plant of Ranunculaceae.Potentilla discolor Bunge is a Potentilla plant of Rosaceae.Pulsatilla chinensis(Bge)Regel is widely used in clinical treatment.However,due to historical and other reasons,Potentilla discolor Bunge is misused as Pulsatilla chinensis(Bge)Regel,leads to the security risks in clinical medication.In addition,traditional identification methods are easily affected by the properties of drugs and environmental factors,so new convenient and accurate methods should be established.Molecular identification could be a good method to identify them from each other precisely.In this study,genomic DNA were extracted from samples of Pulsatilla chinensis(Bge)Regel and Potentilla discolor Bunge and the ITS and the trnL-F region were amplified by PCR.The amplified products were purified and then sequenced directly.Comparison analyses of these sequences were carried out.According to analyses,differences of ITS and trnL-F sequences are found between Pulsatilla chinensis(Bge)Regel and Potentilla discolor Bunge,so direct sequences comparison and PCR-RFLP can be used to identify them from each other? Meanwhile,it also provided a reliable reference for molecular identification of other Chinese traditional medicines.
