Apoptosis Related Genes Bcl-2, The Expression Of Bax In The Scar Tissue In The Bile Duct And Related Research

OBJECTIVE To explore the characteristics of Bax gene and bcl-2 gene expression in benign biliary stricture and normal biliary tissues and its implication.METHODS 20 cases of biliary scars of patients with the benign biliary stricture and 20 cases of normal biliary tissues were collected. The expretion of gax gene and bcl-2 gene in benign biliary stricture and normal biliary tissues by usingimmunhistochemical method and RT-PCR method.RESULTS Light microscopy showed that collageous fiber was arranged in a dense and disorderly way in the biliary scars.RT-PCR:The expretion of Bax mRNA and bcl-2 mRNA in benign biliary stricture was signicantly higher than the expretion.of Bax mRNA and bcl-2 mRNA in normal biliary tissues.(P＜0.05).The radios of expretion of Bax mRNA and bcl-2 mRNA in benign biliary stricture was signicantly higher than the radios of expretion of Bax mRNA and bcl-2 mRNA in normal biliary tissues.(P＜0.05).Immunohistochemical test showed The expretion of gax protein and bcl-2 protein in benign biliary stricture was signicantly higher than the expretion of Bax protein and bcl-2 protein in normal biliary tissues.(P＜0.05).The radios of expretion of Bax protein and bcl-2 protein in benign biliary stricture was signicantly higher than the radios of expretion of Bax mRNA and bcl-2 mRNA in normal biliary tissues. (P＜0.05).CONCLUSIONS The expretion of Bax gene and bcl-2 gene in biliary tissuesmight play synergetic roles in the process of forming benign biliary stricture.Apoptosis is in-hibited in the process of forming benign biliary stricture.The inhibition of apoptosis may be relatively insufficient.Bcl-2/bax is an important regulator of apotosis,and the relative predomination of Bax gene promotes the apop-totic death.
