Preliminary Study Of The Effects Of Colchicine On Mouse Glioblastoma Chemotherapy

Objective To discuss mouse glioblastoma model induced by G422 cell in the brainMethods 20 male mice were separated randomly into a treatment group and a blank control group,each group have 10 mice.The mouse G422 cell were cultivated to 4×10~7/ml in the RPMI 1640 medium,then the G422 cell or natural sodium were micro-injected into mices contex to two groups respectively,we observe the two groups survivalResults Through survival,we discovered that the average survival of group microinjected into by G422 cell was longer than group by natural sodium,there was difference between the treatment group and the blank control group(P＜0.00).Conclusions Through the G422 cells proliferation subcutaneously,the model is reliable Objective To investigate the colchicine chemotherapeutic role on the mice glioblastoma through the coculture G422 cell and colchicine.Methods 30 male mice were separated randomly into a treatment group and a blank control group,each group have 15 mice.The mice G422 cell were cultivated to 1×10~7/ml in the RPMI 1640 medium,then the cell were divided into two parts, each was added equivalently 32ug/ml colchicine or natural sodium respectively,and cultivated in RKI-1002 incubator for 90 minutes,the treatment group was micro-injected to mice contex by mice G422 cell added 32ug/ml colchicine,the blank control group by mice G422 cell added natural sodium.we observe the two groups survivalResults Through survival,we discovered that the average survival of group added 32ug/ml colchicine was longer than group added natural sodium,there was difference between the treatment group and the blank control group(P＜0.05)Conclusion In vitro colchicine suppressed G422 cell proliferation to some degree. Objective:To investigate the chemotherapeutic role of colchicine which is injected into the model of mice intracrianial glioblastomaMethods:50 male mice were micro-injected 4 x10~7/ml mouse G422 cell 10ul into the mice brain contex.After 7 days,the mice tumors were dessection,then the mice were separated randomly into 5 groups,marked A,B,C,D,E,Group A were given 4ug/ml colchicine 10ul into mice brain contex,group B 8ug/ml,group C 16ug/ml,group D 16ug/ml,,group E natural sodium,we observe the mice survival.Result:There was no significant difference in the average survival among the surgical groups and the control group(P＞0.05).Conculsions:In the model of mice G422 cell glioblastoma,colchicine have no effect on survival among the groups by interstitial chemotherapy.
