| Objective:With the rapid development of social and biological sciences,people's living standards get increasingly improved and the aspiring for health and longevity has surpassed any period in the history.Senescence mechanism is being studied further, consequently both western and Traditional Chinese medicine(TCM)theories on senescence are established,making delaying senescence one of the forward topics in present society.Nowdays,western medicine has realized the relationships between senescence and injury of free radical,somatic mutation,abnormal cell apoptosis,mitochondrion DNA mutation,telosome shortage etc.TCM holds that senescence is mainly relevant to deficiency of spleen and stomach,kidney deficiency,phlegm,blood stasis etc.For us,we attach importance on the spleen and stomach and regard that senecence of whole body begins with organ senility,with stomach senility as the leading factor.Combing the above factors and the clinical practice of my tutor,we self make WeiantaiⅡto do some benificial work for the prevention of stomach senility.By establishing the senility animal model with spleen deficiency,this experiment is to study the relationship between spleen deficiency and stomach senility,meanwhile,changes of relevant index are observed in partial gastric mucosa as well as whole body under the influence of WeiantaiⅡin order to probe into the function of WeiantaiⅡon delaying stomach senility.Methods:By hypodermic injection of D-galactose on a two-month-old male rats for 40 days,subacute senescence model were established.Then,by percutaneous absorption of rhubard decocted liquid for 10 days,senescence model with spleen deficiency were established SOD,MDA in the gastric mucosa and serum were detected respectively on the 51th day,6 weeks',12 weeks' treatment by WeiantaiⅡ.Pathological changes in the gastric mucosa as well as organ index of spleen,stomach and thymus gland were also observed.By comparing with blank group,whether WeiantaiⅡexert effect on the stomach senility can be seen.Results:The model group continuously is given D-galactose for 40 days,by percutaneous absorption of rhubard decoctted liquid for 10 days,Rats' hair color being huangbai and scarce,the amount the amount of hair removel,spirit in low with little move,body thinness,lying,Lean stool,Reduce food intake,that present out obviously senescence sign.It proves the senility animal model with spleen deficiency is successfully built with the literature report way consistence.1.comparing with bland group,organ index,SOD in the serum and gastric mucosa had significantly lowered(P＜0.01),with MDA in the serum and gastric mucosa significantly increased(P＜0.01),had significant statistical meaning.2.Comparing with model group,organ index as well as SOD in the serum and gastric mucosa were signicantly increased in the half dose and full dose groups (P＜0.01),while MDA in the serum and gastric mucosa were significantly lowered(P＜0.01),had significant statistical meaning.3.Comparing with half dose group(C1),organ index,SOD in the serum,SOD,MDA in the gastric mucosa,MDA in the serum in the full dose group(D1)had significant meanings,each with(P＜0.01),(P＜0.05),(P＜0.01),(P＜0.01),It prove that the full dose group is superior to the half dose group.However,organ index,SOD,MDA in the serum in the half dose group(C2)on the therapeutic time had no statistical meaning(P＞0.05),while SOD and MDA in the gastric mucosa had (P＜0.01),It's clear that improvement trend to some markets of half dose group in 12 weeks acting on is superior to 6 weeks,but the relation don't obvisously show.4.Comparing with full dose group(D1),full dose group(D2)had no significant differences in the organ index.In the therapeutic time,SOD in the gastric mucosa had no significant meaning(P＞0.05),while SOD in the serum had.(P＜0.05).It's clear that improvement trend to some markets of full dose group in 12 weeks acting on is superior to 6 weeks,but the relation don't obvisously show.5.Histopathological tissue section of gastric mucosa showed that WeiantaiⅡcan repair the gastric mucosa of senility rats and promote the regeneration of gland by comparing the difference in gastric morphology between treatment group and model group.Conclusion:This experiment has testified that WeiantaiⅡpossess the function of anti-oxidation,clearing free radical,enhancing immune ability,reparing the injured gastric mucosa,promoting the regenerationof inherent gland etc.It shows that WeiantaiⅡcan delay the senecence of stomach and whole body in a certain way,which provide therapeutic clue and drug selection for clinical paractice. |
