This thesis mainly contains five chapters. The first chapter discussed the chemical constituents for volatile oil of Miao Ethnomedicine Herminium bulleyi(Rolfe)Tang et Wang by three methods. The second chapter dilated the isolation and identification of chemical compounds from Herminium bulleyi(Rolfe)Tang et Wang. In the third chapter, we extracted the polysaccharide from Herminium bulleyi(Rolfe)Tang et Wang, and determined its contents.The fourth chapter discussed the toxicity test a series of immunopharmacological tests(avoirdupois and the weight and index immune organs, serum hemolysin test) to normal ICR mice for the polysaccharide of Herminium bulleyi(Rolfe)Tang et Wang. In the last chapter ,we have a study of literature about arethusa. The results of the primary study on the chemical constituents and pharmacological activity from Herminium bulleyi(Rolfe)Tang et Wang were reported in this thesis.1.The volatile oil of Herminium bulleyi(Rolfe)Tang et Wang was extracted by CO2-supercritical fluid extraction and steam distillation and ether extraction, and their chemical constituents were analyzed and identified by GC-MS.Totally 46 components were detected, among them 32 were identified from the volatile oil extracted by SFE-CO2.Its main components are palmitic acid (52.16%), camphor (8.30%), hexadecanoic acid ethyl ester (7.27%), oleum eucalypti (2.97%), dissobutyl phthalate (2.51%), borneol(1.32%); Totally 59 components were detected, among them 33 were identified from the volatile oil extracted by SD. Its main components are palmitic acid (26.66%), linoleic acid (24.23%); Totally 96 components were detected, among them 50 were identified from the volatile oil extracted by ether, its primarily were some alkane compounds and their derivates.2 . From above, nine compounds were isolated and purified by column chromatography on silica gel and RP-18 and sephadex LH-20 from the tuber of Herminium bulleyi(Rolfe)Tang et Wang. Based on the spectrum analysis of their 1H-NMR, 13C-NMR and MS datas, structurers of six compounds were identified, respectively as Palmitic acid (2);9,19-cyclo-9β-lanost-24-en-3-ol (3);β-sitosterol (4); glycerol monopalmitate (5); Monostearin (7); 2,2-dimethyl-5-methoxy-7-[2-(3''-hy- droxyphenyl)ethyl]chromene (8).The above compounds were isolated from this plant for the first time, among them, compound 8 was a new compound.3.Determination the content of polysaccharide from Herminium bulleyi(Rolfe)Tang et Wang by the method of phenol-oil of vitriol for the first time, its content is 58.5%.4.The toxicity test a series of immunopharmacological tests(avoirdupois and the weight and index immune organs, serum hemolysin test) to the normal ICR mice for the polysaccharide of Herminium bulleyi(Rolfe)Tang et Wang for the first time.The test result presented that the polysaccharide of Herminium bulleyi(Rolfe)Tang et Wang could remarkable increased the weight of spleen of the ICR mice, advanced the generation of antibody, combine with the variety of avoirdupois, we could deem that the polysaccharide of Herminium bulleyi(Rolfe)Tang et Wang to the humoral immunity of ICR mice have some reinforcing effect. It indicated the polysaccharide could enhance the specific immune function on mice.This thesis took the study on the chemical contstituents and pharmacological activity of Herminium bulleyi(Rolfe)Tang et Wang for the first time. For its further exploitation and utilization established theoretical basis as well as for the study of Orchidaceae chemotaxonomy.
