Objective To establish an animal model similar to the metabolic abnormalities of human type 2 diabetes mellitus with insulin resistance.Discuss the relation between the model and blood-fat,antioxygen parameters.Methods Eight-week-old SD male rats(n=120)were divided into two groups: control group and model group.The model group male SD rats(n=90)were fed with high-fatty diet that contained unsaturated fatty acid for 4 weeks and then streptozotion (STZ,one-step,25mg/kg)was injected intraperitoneally,then streptozotion(STZ,40 mg/kg)was injected intraperitoneally again in the next 4 weeks to result in insulin secretion defect and hyperglycemia.The control group(n=30)were fed with normal diet.Blood-fat was detected by biochemistry equipments.According to kits' directions determin GSH-Px.Results Biochemical parameters proved that the two-step treatment methods could develop the T2DM rat model.LDL was 0.75±0.32mmol/L,TC was 1.43±1.07 mmol/L,TG was 0.79,1.59,2.04 mmol/L,GHb was 6.71±1.37%of this T2DM group were all higher compared with control group,and GSH-Px was lower comared with control group.Conclusion Type 2 diabetes mellitus animal model is successfully reproduced by using the high fatty diet plus STZ injection.In the T2DM group,the successful rate of execution was 72%,the death rate of execution was 33%;It may provide a perfect animal model for investigating type 2 diabetic mechanism and pharmacology,also be used to research insulin resistance and related diseases of human beings.LDL,TC,TG and GHb of this model group were step up obviously,GSH-Px activity was to lower. Objective Intracerebral infarction(CI)induced by diabetes is an emergent disease endangering the people's life and work because of its high mortality and serious disability.The most important pathological changes after CI are the ischemic itself and the secondary edema,which causes herniation-related death and severe neurological deficits.Although lots of improvements have been made on the prevention and treatment of CI,the mechanism of CI induced by diabetes formation and secondary neuron injury are not very clear now.So the researches on CI become the focus of clinical and primary studies.Up to now,there is no one kind of mechanisms to explain the damage theorem after cerebral ischemia.Objective to establish the model of focal cerebral ischemia affiliate diabetes with favorable stability,and investigate the syndrome of neurological severity,the volume of cerebral infarction and the pathological changes after different ischemic time.Methods Making middle cerebral artery ischemic model in SD rats.10 SD rats were randomly divided 2 groups.Rats in group A1 were sham-operated for control study. Unilateral middle cerebral artery occlusion(MCAO)was performed with improved thread in group B1;30 SD rats were randomly divided 2 groups.Rats in group A were sham-operated for control study which were diabetes rats.Unilateral middle cerebral artery occlusion(MCAO)was performed with improved thread in group B which were diabetes rats.Neurological severity score(NSS),triphenyl tetrazolium chloride (TTC)staining and HE staining were adopted to observe dynamically the syndrome of neurological severity,the volume of cerebral infarction and the pathological changes. The area of infract and neurological deficit were monitored,the successful rate of execution,neurological deficit examination,infarct area of brain and the model stabilization of three groups were compared.Results In the group A1,the successful rate of execution was 100%.In the model B1,the successful rate of execution was 66.7%.In the model A2,the successful rate of execution was 75%,neurological deficit scoring of 3-4 grade 75%and infarct area of brain 55.23±9.59 mm~3 respectively.In the model B2,the successful rate of execution was 53.3%,neurological deficit scoring of 3-4 grade 60%and infarct area of brain 54.33±6.39 mm~3 respectively.All rats had different neurological deficit scores at different time points after CI.The limbs of rats was hemiplegia after operation.The behavior changes were no difference between the CI group and the Sham group at 6 hours after operation.Possibly the anesthetic and operation injury lead to the result. The sham group A1 and B1 behavior change normal at 24h.Conclusion The model of MCAO is favorable stability.The model in this study is reversal,and ischemic repeffusion research can be made on the basis of it.It is applicable to the acute and chronic empirical study.After CI,the changes of neurological behavior scores and the brain water content were consistent,which suggested that brain edema was the most important cause leading to neurological function deficit after CI.
