Objective: To investigate effect of Prokineticin2 on neural stem cell migration and differentiation in vitro and brain of MCAo rat.Methods:The advantage of serum free and clone culturing technology is performed to isolate,culture and passege neural stem cells from neonatal rat, immunohistochemistry used for identification of nestin pigmentation.Neural stem cells be placed in the PK2 scattered around the source of proliferation,PK2 proliferation source be made of 5% agar and PK2. According to the different doses of PK2,groups are as follows:1nmolPK2,30nmol PK2,60nmol PK2,control group without PK2. Training in basic culture medium, basic culture medium containing 0.3% agar the DMEM/F12 (withdrawal bFGF). Cultured in the DMEM/F12 cell culture medium, culture medium containing 3% agar and removed bFGF. Continuous observe neural stem cells in 14 d. Screening of a neural stem cell migration induced by the best role PK2 dose. Thread of the Wistar rats into middle cerebral artery occlusion (MCAO) model. MCAO model rats were randomly divided into 3 groups: model group, control group (simply transplanted NSCs), PK2 group (PK2 and NSCs common transplant), 24 / group. Using three-dimensional positioning, after BrdU labeling of the NSCs transplanted into rat MCAO model to the right lateral ventricle, the best dose PK2 injected in the right lesion area of MCAO model rats. Using immunofluorescence method to observed that neural stem cells of moved to the lesions differentiate into neurons .Results:â‘ In vitro studies showed, compared with the control group, the different doses of Prokineticin2 have induced migration of neural stem cells, and 1 nmol Prokineticin2 induced migration of the most obvious role, there are statistically significant differences(P<0.01).â‘¡Transplantation of neural stem cells in 1d, compared with model group in SVZ, the BrdU-positive neural stem cells have markedly increased in simple neural stem cell transplantation group and PK2 groups, there are statistically differences (p <0.05), PK2 group compared with the control group, the number of BrdU-positive cells in SVZ there are no significant difference (p> 0.05); In the lesion area of every group, the number of BrdU-positive cells are no significant difference (p> 0.05). In 3,7,14 d, PK2 group in the SVZ BrdU-positive cells was higher than the model group and the control group, there are statistically differences (p <0.05). PK2 group in the lesion area BrdU-positive cells than the model group and the control group,and differentiate into neurons ,there are statistically differences (p <0.05).Conclusion:PK2 may induces the migration of neural stem cells in vitro, 1 nmol PK2 is the effect in most obvious. And PK2 can be induces the migration of neural stem cells that transplanted into the MCAO rat brain,and PK2 promote the differentiation of neural stem cells to nerons.
