Objective:To detect the expression of spleen tyrosine kinase(Syk) in benign and malignant endometrium,and to evaluate the effects of the Syk expression in endometrial carcinoma on tumor growth and metastasis,and the correlation between the Syk expression and the clinical features of the tumor.Methods:Forty cases of endometrial carcinoma tissues samples,twelve cases of atypical hyperplasia tissues,ten cases of endometrial simple hyperplasia and endometrial complex hyperplasia tissues and ten cases of normal endometrial tissues were collected.PV6000 immunohistochemistry method was applied to detect the expressions of Syk protein in normal endometrial tissues and various endometrial changes tissues specimens.Results:The positive staining of Syk protein was located in cytoplasm.The positive rates of Syk protein were 100%(10/10),90%(9/10),75%(9/12),42.5%(17/40) in normal endometrial tissue, endometrial simple hyperplasia and endometrial complex hyperplasia tissues,atypical hyperplasia tissues and endometrial carcinoma tissues respectively.The rate of positive Syk expression in endometrial carcinoma reduced significantly and was lower significantly than that in normal endometrial tissues,endometrial simple hyperplasia and endometrial complex hyperplasia tissues and atypical hyperplasia tissues separately(P<0.05);Only three endometrial carcinoma tissues in nineteen patients with lymph node metastasis had the Syk protein expression,fourteen endometrial carcinoma tissues in twenty-one patients without lymph node metastasis had the Syk protein expression.The Syk protein expression was negatively correlated to lymph node metastasis(p<0.05).Expressions of Syk ofⅢ+Ⅳstages were lower than those ofⅠ+Ⅱstages of endometrial carcinoma,but the expressions of Syk were not related to histological grade and depth of infiltrating muscular layer(P>0.05).Conclusion:The expression of Syk is on descending trend in evolution process of endometrium from binign to malignant.The reduced or lost expressions of Syk may play an important role in suppressing growth and metastasis of endometrial carcinoma,and indicate that Syk may be a candidate tumor suppressor of this kind of carcinoma,and the expression of Syk is correlated with malignant chang of endometrium.
