Objective: To explore the Protective function and mechanism of Mai Tong Fang on Diabetic Rats with Erectile Dysfunction. Seek evidence for clinic use of the formula from experiment.Method: The 50 SPF grade SD male rats were randomly divided into 2 groups: Experimental group and Normal group. The Experimental group was injected intraperitoneally with STZ and the Normal group with Sodium Chloride for comparison. The rats were fed with normal food for 15 weeks after the models of Diabetic Rats were successful. 15 weeks later, the 31 Diabetic Rats were divided randomly into to 2 groups according to the FPG: Model group and Treatment group. Decoction (equal to 18.125g/kg of Mai Tong Fang compound herbs) for each rat in Treatment group. The same volum of distilled water for Model group and Normal group. 15 days later, we sheared the tails of rats for the peripheral whole blood to measure the FPG and the content of SOD; and got the tissues of penis to measure the expression of iNOS, nNOS and NF-κB, and the ratio of contents of fibroblast and smooth muscle cell.Results: Compared with the Model group, the Treatment group was observed with a better general state of health, but worse than Normal group. The FPG of Model group is higher than Treatment group and Normal group, and Treatment group is higher than Normal group. There're statistical significances of these differences (P<0.05). About the content of SOD of whole blood and the positive expression of NF-κB: Treatment group's and Model group's are lower than Normal group's (P<0.05). There's statistical significance of these differences. But there'satistical significant difference between Treatment group's and Model group's (P>0.05). About the positive expression of nNOS: Treatment group's and Model group's are lower than Normal group's, Treatment's is higher than Model group's, there're statistical significances of these differences (P<0.05). About the positive expression of iNOS: Treatment group's is lower than Model group's, Normal group's is lower than Model group's, there're statistical significances of these differences (P<0.05). But there's no statistical significant difference between Treatment group's and Normal group's (P>0.05). About the ratio of contents of fibroblast and smooth muscle cell: Model group's is higher than Normal group's, Treatment group's is higher than Normal group's, there're statistical significances of these differences (P<0.05). But there's no statistical significant difference between Treatment group's and Normal group's (P>0.05). Conclusion: Mai Tong Fang can improve the hyperglycaemia state of DMED Rat, increase the positive expression of nNOS of tissues, and cut down the expressions of iNOS and NF-κB of tissues. It means that Mai Tong Fang can adjust BG, anti-oxidation, and anti-apoptosis. The mechanism of improving the DM rat's ED could be concerned with the difference of above.
