Gynura segetum is the dry root tuber of Compositae plant Gynura segetum(Lour.) Merr.,recorded in the drug standard of ministry of Public Health of the People's Republic of China(Chinese crude drug).The efficacies include hematischesis, analgesia and detumescence by detoxification.The circs of modern study of pharmacology indicate that Gynura segetum can assuredly stop bleeding,so it possesses well foreground of exploitation.In view of the actuality that the basic research on Gynura segetum was fall behind and lack of perfect method of quality control,the subject is to screen out the active fractions and components for hemostasis from Gynura segetum by the pharmacodynamical experiments in vivo and in vitro.Basing these researches,we studied the fingerprint of the effective fraction and the method of quality control of Gynura segetum.By the hemostatic experiment of the marginal ear AtV of rabbit and the experimemt of testing CT,PT,APTT,FIB,the research screen out the active fraction which was got from Gynura segetum through the systemic solvent distillation method. The result shows that Acetic Ether extracts is the active fraction for hemostasis,which can obviously shortern the hemostatic time of the marginal ear AtV of rabbit,so did CT,PT,APTT,FIB.At the same time,it can promote the increase of FIB, moreover the activity is stronger than Yunnan white powder.The Acetic Ether extracts was separated and purified by silica column and Rp-C18 column laminar analysis,then four compounds were isolated,and three of them were identified as Senecionine,Seneciphylline and Senecionine N-oxide by spectroscopy methods such as UV,1H-NMR and 13C-NMR.All of them were confirmed as the effiective compounds which have significant activity on blood clotting.Studied on the fingerprint of the effective fraction---Acetic Ether extracts of Gynura segetum by RP-HPLC,and established the standard HPLC-FPC.The result showed that all samples had the similar common-characteristic in the fingerprint chromatogram,and the similarity was above 0.900.This method using for controlling the quality of the effective fraction is stable,reliable and has good repeatability as well as applicability.At the same time,it may supply the techincal conduct for the construction of GAP base.In addition,we found out a great difference between the rhizoma and the aerial parts of Gynura segetum by the contradistinctive research of their fingerprint.The result can offer the techincal support for the quality monitoring of the product on the market,and then it also may provide credible information for the purchases of medical material.In this research,the quality specification of Gynura segetum was improved and consummated systematically.Referring to the methods of China Pharmacopoeia (2005 edition Spl.),we tested moisture content,heavy metals,As,ash and ethanol-soluble extraction.For the first time,the subject took Senecionine as reference substance to setup the identification means by TLC,and two assaying methods for total TA and Senecionine were established by UV-VIS and HPLC, furthermore determined three batches of medicinal materials.On account of its convenience,sensitiveness,veracity and good repeatability,the method is propitious to control the quality of Gynura segetum.Through the research of the subject,we have a systematical study on the potential basis of Gynura segetum,and screened out the effective fraction and three effective constituents for hematischesis.Furthermore established the standard HPLC-FPC of the effective fraction for hemostasis.Meanwhile,we setup the scientific and consummate quality specification of Gynura segetum,according to which we can not only control the quality of medicinal materials availably,but also lay a foundation for the research of new-style hemostatic for munition.
