Objective:To observe the distribution laws of immune substances in lung and other organs of rats,of which the two Feishu points are given moxibustion and then with intraperitoneal injection of LPS.To explore the laws and mechanisms of the relativity between acupoints and visceras.And observing the effect on HPA axis by Feishu moxibustion,and also the correlation betwwen HPA axis and immune function.Methods:Thirty two SD rats were randomly divided into four groups,namely blank group, LPS group,Feishu LPS group,Ganshu LPS group,n=8.Rats acute inflammatory response model were made by intraperitoneal injection of 2mg/kg of LPS solution to rats.In Feishu LPS group and liver LPS group,rats hairs in the Feishu and Ganshu area,should be respectively sheared,bilateral acupoints should be given moxa cone moxibustion,and changing cones when rats twist,5 Zhuang each tiom, 1 times daily for 10 days.In the blank group and the LPS group,rats hairs in the Feishu area should be sheared,and with the same catching and no moxibustion for the former ten days.The day of the eleventh day,rats of the blank group were directiy anesthetized and drawed materials,and rats of the remaining three groups should get intraperitoneal injection of LPS 2mg/kg,and be anesthetized after 6h.To assay the required tissue homogenate supernatant and serum TNF-a, IL-2,TGF-β,sICAM-1 and HPA-axis content by radioimmunoassay and enzyme-linked immunosorbent assay methods,also calculte the organ index of different organs and tissues.Results:1.Intraperitoneal injection of LPS impact on spleen and thymus.Intra-group comparison,the TNF-a content in spleen and thymus of blank group is of the highest content,and have significant difference with the serum concentration(P<0.05);TNF-a content in spleen and thymus of LPS group is in the last two sequence,and have extremely significant difference with the kidney concentration.Comparison among groups,TNF-a content of spleen,thymus in LPS group is significantly lower than the blank group(P<0.05).The spleen index of LPS group is significantly higher than that of blank group(P<0.05);the spleen index of Feishu LPS group and Ganshu LPS group is significantly higher than that of blank group(P<0.01);the thymus index of Ganshu LPS group is significantly higher than that of LPS group(P<0.05).2.The Feishu moxibustion effect on immune substances in lung.Compared to blank group,TNF-a content in lung of LPS group has the downward trend.TNF-a content in lung of Feishu group is significantly lower than that in blank group,and has the downward trend comparing to LPS group.After sorting,compring to LPS group,the place of TNF-a content in lung of Feishu group moved to the last place,and has very significant difference compared to the TNF-a content in kidney,which is in the fourth place.Compared to blank group,IL-2 content in lung of LPS group has the downward trend.IL-2 content in lung of Feishugroup is significantly lower than that in blank group,and has the downward trend comparing to LPS group.After sorting,compring to LPS group,the place of IL-2 content in lung of Feishu group moved back,and has no statistical significance compared to the IL-2 content in other organs.Compared to blank group,TGF-βcontent in lung of LPS group has the downward trend.TGF-βcontent in lung of Feishu group has upward trend compared to blank group and LPS group.After sorting,compring to LPS group,the place of TGF-βcontent in lung of Feishu group moved back one place,and has no statistical significance compared to the TGF-βcontent in liver which is in the first place.Compared to blank group,sICAM-1 content in lung of LPS group has the downward trend,sICAM-1 content in lung of Feishu group has upward trend compared to blank group and LPS group.After sorting,compring to LPS group,the place of sICAM-1 content in lung of Feishu group moved forward one place.3.The Feishu moxibustion effect on immune substances in lung and related other organs.Compring to LPS group after sorting,the place of TNF-a content in kidney of Feishu group move back,and that in heart move to the first place.Comparing to blank group and LPS group,TNF-a content in kidney of Feishu group has the downward trend.Comparing to LPS group,TNF-a content in thymus,liver,serum,and heart of Feishu group increase,and TNF-a content in lung,intestine,spleen,and kidney decrease,and has no statistical significance.Compring to LPS group after sorting,the place of IL-2 content in heart and liver of the Feishu group is in the first and second place,the place of IL-2 content in lung and kindey move back,but there is no statistical significance between different organs.Comparing to LPS group,IL-21 content in liver,lung,heart,intestine,and serum of Feishu group increase,and IL-2 content in serum of Feishu group is significantly higher than that of LPS group. Compring to LPS group after sorting,the place of TGF-βcontent in liver of Feishu group move forward to the first place,and is significantly and extremely significantly higher than that in heart and serum.Comparing to LPS group,TGF-βcontent in all the organs and tissues of Feishu group has the upward trend.Compring to LPS group after sorting,the sICAM-1 content in liver of Feishu group increase and is in the first place,and is significantly higher than that in serum.Comparing to LPS group,sICAM-1 content in kidney,heart,and serum of Feishu group has the downward trend,sICAM-1 content in intestine,spleen,liver,and lung increase,and sICAM-1 content in spleen is significant]y higher than that in LPS group.4.The Feishu moxibustion effect on immune substances in liver.Compared to blank group,TNF-a content in liver of LPS group and Ganshu group has the upward trend.Comparing to LPS group,TNF-a content in liver of Ganshu group has the downward trend,and the place of TNF-a content in liver of Ganshu. group move bace two place.Compared to blank group,IL-2 content in liver of LPS group and Ganshu group has the upward trend.Comparing to LPS group,IL-2 content in liver of Ganshu group has the upward trend,and the place of IL-2 content in liver of Ganshu group has no change.Compared to blank group,TGF-βcontent in liver of LPS group and Ganshu group has the downward trend.Comparing to LPS group,TGF-βcontent in liver of Ganshu group has the downward trend,and the place of TGF-βcontent in liver of Ganshu group move back one place and the TGF-βcontent in liver had no significantly difference with that in other organs of Ganshu group.Compared to blank group,sICAM-1 content in liver of LPS group has the upward trend,and sICAM-1 content in liver of Ganshu group has downward trend.Comparing to LPS group,sICAM-1 content in liver of Ganshu group has the downward trend,and the place of sICAM-1 content in liver of Ganshu group move bace one place.5.The Ganshu moxibustion effect on immune substances in liver and related other organs.Compring to LPS group after sorting,the place of TNF-a content in liver of Ganshu group move back,the place of TNF-a content in heart move forward.The place of TNF-a content in iung and intestine of Ganshu group move back too,and is significantly lower than that in kidney.Comparing to LPS group,TNF-a content in lung,intestine,spleen,liver and kidney of Ganshu group has the downward trend, TNF-a content in thymus,heart,and serum increase,but has no statistical significance.Compring to LPS group after sorting,the place of IL-2 content in liver of Ganshu group has no change,the place of IL-2 content in lung move back two place. Comparing to LPS group,IL-2 content in liver,intestine,spleen, kidney,thymus,and serum of Ganshu group has the upward trend,IL-2 content in lung and heart has the downward trend,but has no statistical significance.Compring to LPS group after sorting,the place of TGF-βcontent in liver of Ganshu group has no change,and TGF-βcontent in liver of Ganshu group increase and is significantly and extremely significantly higher than that in kidney,heart,and serum.The place of TGF-βcontent in liver of Ganshu group move back one place.Comparing to LPS group,TGF-βcontent in liver and serum of Ganshu group decrease,TGF-βcontent in lung,spleen,kidney,and heart of Ganshu group increase,but with no statistical significance.Compring to LPS group after sorting,the place of sICAM-1 content in lung of Ganshu group move forward and is in the second place,and the sICAM-1 content in lung of Ganshu group is significantly higher than that in liver and serum.Comparing to LPS group,sICAM-1 content in liver,kidney,heart and serum of Ganshu group decrease,sICAM-1 content in intestine,spleen,and lung of Ganshu group increase,but with no statistical significance.6.The effect on HPA axis by Feishu moxibustion and intraperitoneal injection of LPS.The CRH content in LPS group is significantly lower than that in blank group,and ACTH in pituitary,COR in adrenal gland of LPS group has the downward trend comparing to blank group.The CRH content in Feishu LPS group is extremely significantly higher than LPS group,and has the downward trend comparing to blank group;The ACTH content in pituitary of Feishu LPS group has the downward trend comparing to LPS group,and is significantly lower than LPS group;The COR content in adrenal gland of Feishu LPS group has the upward trend comparing to blank group and LPS group.CRH,ACTH in pituitary,COR in adrenal gland of Ganshu LPS group has the upward trend comparing to LPS group and blank group,and CRH content in Ganshu LPS group is significantly higher than LPS group.7.The correlation between HPA axis and the immune function of rats.Comparing to blank group,the content of CRH,ACTH in pituitary in Feishu LPS group has downward trend and the content of COR in adrenal gland in Feishu LPS group has upward trend;the content of CRH,ACTH in pituitary,COR in adrenal gland in Ganshu LPS group all have upward trend.To anlysi s the correlation between COR in adrenal gland,COR in serum and every immune substances,there is no correlation between COR in adrenal gland,COR in serum and immune substances in LPS group.Few immune substances have positive correlation with COR in adrenal gland,COR in serum in Feishu LPS group and Ganshu LPS group. Conclusion:1.Feishu with lung,and Ganshu with liver are closely associated with each other.Feishu with moxibustion can lighten the inflammation of lung by decreasing the content of TNF-α,IL-2 and increase the content of TGF-β,sICAM-1 in lung.And Ganshu with moxibustion can lighten the inflammation of liver by decreasing the content of TNF-a,TGF-β,sICAM-1 in liver.2.Acupoints are associated with visceras,and the relationship is one acupoints related with more visceras.Regulation law to TNF-αby Feishu with moxibustion,and regulation law to sICAM-1 by Ganshu with moxibustion are coincident with the generation-restriction law in five elements.So we consider that the mechanism of the relativity between acupoints and visceras is the law of regulating different cytokine between visceras,and the laws are corresponding with the generation-restriction law in five elements.3.Of course there are still some cytokine don't show the same law,we think the five elements law is just one of the laws in the relationship between visceras,there are many other laws like the relationship of the function,or maybe the results have connection with the time,materials,and so on.4.The HPA axis has the trend of beening surpressed after intraperitoneal injection of LPS,and Feishu,Ganshu with moxibusition both have the trend of preventing the surpressing trend,or maybe the moxibustion and the injection are stress stimulation,and the HPA axis has the trend of being excited,HPA axis of rats in Feishu LPS group and Ganshu LPS group has positive correlation with the immune function of rats.
