Objective:To explore the effects of acupuncture on uricopoiesis metabolic enzymes activities of Hyperuricemia(HUA)rats,and to elucidate the possible mechanism of acupuncture at"ganshu"(BL18)and "taichong"(LR3)for the prevention and treatment of HUA.Methods:32Wistar male rats were randomly divided into blank,model,shenshu and ganshu groups.The blank group was given 10ml/kg of normal saline every day,and the rest groups were given potassium oxazidate 2g/(kg.d)as 10ml/kg to replicate the HUA rats.Shenshu and ganshu groups were respectively given the intervention of shenshu,taixi point,ganshu and taixi point,20min/time,1 time per day,6 times of rest for one day,lasting 3 weeks.The content of serum uric acid(SUA)in rats was detected by automatic biochemical analyzer,the activity of XOD and adenosine deaminase(ADA)in liver homogenate of rats was observed by colorimetry,the positive expression of XOD was detected by immunohistochemistry(IHC),and the activity of guanine deaminase(GD)was detected by Elisa.Results:Cmpared with the blank group,the activity of XOD,ADA and GDA in the model group was significantly increased(p<0.05)and the relative expression level of XOD positive was increased(p<0.01).Compared with the model group,the activity of XOD and ADA in shenshu group and ganshu group was decreased(p<0.01),the relative expression level of XOD positive was significantly decreased(p<0.01),and the GD activity in ganshu group was decreased(p<0.01).Compared with shenshu group,the relative expression level of XOD positive in ganshu group was decreased(P<0.05).Conclusion:Acupuncture on ganshu and taichong acupoints can inhibit the activity of XOD,ADA and GD in HUA rat models,and may directly affect the metabolic pathway of purine decomposition,inhibit the activity of uric acid metabolic enzymes,and affect the production of uric acid.
