Objective To detect hypermethylation status of the 5'CpG island locating in the promoter region of hMLH1(mismatch repair genes,MMR)gene and the expression level of their mRNA in gastric carcinoma and analyze the relationship between DNA methylation status and the tumorigenic process of gastric carcinoma.Methods Methylation of the hMLH1 promoter was examined by methylation specific PCR(MSP) in 60 gastric carcinomas and 60 adjacent normal tissue,and then,the mRNA expression patterns of hMLH1 gene was detected by RT-PCR method.Results The incidence of the hMLH1 promoter methylation was 30 %(18/60),No methylation of hMLH1 promoter was found in 60 cases of adjacent normal tissue.Methylation did not correlate with clinicopathological characteristics of gastric carcinoma.17 of 21(81%) hMLH1 mRNA negative tumors exhibited prominent methylation,compared with 1 of 39(3%) tumors with positive expression of hMLH1 mRNA.Conclusion hMLH1 methylation was associated with the absence of hMLH1 mRNA expression and might be the underlying cause of mismatch repair gene defects in gastric carcinoma.Promoter methylation of the hMLH1 involved in tumorigenic process of gastric carcinoma.
