Objective:Study the expression of chemokine in nasal mucosa and lung tissue on the base of the model of Allergic Rhinitis(AR) in Rat.Reveal the pathogenic mechanism and pathologic process of allergic rhinitis through observing the effect of yu ping feng powder on chemokine in Allergic Rhinitis in Rat.Method:Forty-eight SD female rats were randomly divided into two groups:Normal group(10 rats),model group(30 rats). In model group,OV were injected into the abdomen cavity and dripped into the nasal cavity to set up AR model.Then,30 rats in model group were randomly divided into AR group,TCM group and Western Medicine group equally.TCM group were given decoction of yu ping feng powder.Western Medicine group were given emulsion of Loratadine Tablets(CLARITYNE).The pathologic samples of nasal mucosa and lung tissue were stained with HE.The expression of Eotaxin and RANTES in nasal mucosa and lung was detected by immunohistochemistry.Result:1.Pathological observation of the nasal septum mucosa and lung tissue:comparison of EOS positive rate of the nasal septummucosaand lung tissue,western medicine group had no statistical significance with TCM group(P>0.05),but both had evident difference with AR group and normal group(P<0.01). 2.Test of IgE:western medicine group had no statistical significance with TCM group(P>0.05),but both had evident difference with AR group and normal group(P<0.01).3.Test of chemokine:Count the positive cell number of chemokine under photo-microscope,western medicine group had no statistical significance with TCM group(P>0.05),but both had significant difference with AR group and normal group(P<0.01).4.Analysis of the relativity:the EOS in nasal septum mucosa and in lung tissue evidently presented positive correlation.With the statistics analysis of expression quantity in septum mucosa, the count of EOS and chemokine in nasal septum mucosa correlated significantly.Conclusion:1.High expression of Eotaxin and RANTES in nasal mucosa of rats with AR,which has the same location as infiltration of eosinophil,It also indicated Eotaxin and RANTES played an important role in pathological process of AR.2.There was similar the pathological changes of the nasal septum mucosa and lung tissue of AR of rats.High expression of Eotaxin and RANTES in lung tissue of AR of rats,revealed there was distinct consistency and correlation in respect of inflammatory reaction of the upper and lower respiratory tracts. It afforded theoretical basis to the research on the lower respiratory tract the research on the upper respiratory tract.3.By restraining the expression of chemokine in nasal mucosa and in lung tissue of rats with AR,Yu Ping Feng powder can decrease the infliltration of EOS in nasal mucosa and in lung tissue of rats.
