This issue is about the biotransformation of active ingredients of TCM by PCC7120,to investigate its biotransformation characteristics and its biotransformation capacity.First of all,it is necessary to establish a stable culturing system for PCC7120 Cyanobacteria,including separation and purification of cyanobacteria,liquid and solid cultivation of algae species,preservation, determination of the growth curve and so on.Comparing two methods of cyanobacteria cell disruption,that is, repeated freeze-thaw and ultrasonic fragmentation,the result show that repeated freeze-thaw is suitable for our experiment because it can deal with large-scale sample.By studying effects of different organic solvents to PCC7120,it shows that Methanol and DMSO in low concertration have a little cytotoxicity while chloroform is very toxic to the cell.After purificated,Rutin has been detected by TLC and HPLC.Its purity is above 98%and meet the needs for biotransformation.After added into BG11+N Culture medium,rutin is vulnerable to degradation in the pH = 7.2,and when the pH = 5.5,the existence of rutin can be stabilized.So it is difficult to control rutin biotransformation by PCC7120, and no new product has been detected by TLC and HPLC.Another two compounds are also added in PCC7120,and in preliminary study,they have no significant transformation.
