| Background&Objective Decorin is a member of proteoglycans families,,whose biologic effect is very active.Decorin is a kind of anti-fibrosis protein which is playing its role in the regulation of extracellular matrix,anti-apoptosis,anti-inflammation and anti-adherence.Decorin can neutralize and inactivate transform growth factor-β1 (TGF-β1).Decorin is the most abundant proteoglycan in normal dermis,while it is little in hypertrophic scar tissue.To observe the effect of decorin on inhibiting hypertrophic scar of rabbit ears.Methods 8 New Zealand white rabbits were used to estabilish the model of hypertrophic scar of rabbit ears,which was randomly divided into 2 groups.20 days after surgery,the hypertrophic scar were injected intralesionally with saline(group A),decorin (group B).30 days after surgery injected again.The scars were harvested 20 days after the last injection for measurement of scar elevation index(SEI).The numerical density on area (NA) of fibroblasts was calculated with HE-stained sections.The expression of type 1 collagen protein and TGF-β1 were addessed by immunohistochemistry.The expression of type 1 pro-collagen and TGF-β1 mRNA were assessed by reverse transcription polymerase chain reaction(RT-PCR).Results The injured area epidermal keratinization of experimental group is better,the scar is more soft,the luster is closer with the periphery normal skin and the scar proliferation degree is reduced compare with the control group.The expression of SEI, NA of fibroblast,type 1 pro-collagen mRNA and TGF-β1 mRNA,type 1 collagen protein and TGF-β1 decreased in group B,,which showed statistical difference compared with groups A(P<0.05).Conclusuion The results of animal experiments suggest that decorin is able to inhibit the proliferation process of hypertrophic scar in rabbit ears and to remarkably decrease the degree of fibrosis in the scar.Decorin would become a new clinical treatment of hypertrophic scar in the future. |
