PurposeTo observe the effect of electro-acupuncture simulating at "Neiguan" on expression of cAMP-PKA in the signal transduction through the experiment of tachyar rhythunia.To explore the meanchism of G protein taking part in the electro-acupuncturing single,explaning electro-acupuncture at "Neiguan" how to influence heart rate upset rats in the point of molecular biology and promote a safe way curing tachyar rhythunia.Material and methods40 wistar rats were randomly divided into blank control group(group A,n=10), model group(group B,n=10),"Neiguan" group(group C,n=10),non-acupoint group(group D,n=10).Injecting 20%urethane in the abdoem after weighing,brady arrhythmia model was established using sublingual verapamil intravenous (5mg/Kg).observing the change of cardiac rhythm by ECG as the single of success.After anaesthesia,the dorsal-bit fixed,connected multimedia MS2000 Record of biological signal analysis system to observe the ECG records.Giving acupuncture stimulate to the model group.After 20 minutes,recored the ECG of blank control group;the model group were injected corcilox agents to make model of brady rhythmia,and 8 minutes later record ECG;the other two electro-acupuncture group make sample 5 minutes later the electro-acupuncturing stimulate.Observe the content of cAMP and the content of PKA in the muscle of rat heart.Results1.Each group of ECG:The heart Rates of model group slowed down significantly, after the treatment of electro-acupuncturing at "Neiguan",the heart Rates of brady arrhythmia rats had Recovered.2.The content of cAMP:The content of cAMP of model group was greatly higher than blank control group and "Neiguan" group(P<0.05);The content of cAMP of non-acupoint group was greatly higher than blank control group and "Neiguan" group(P<0.05).3.The content of PKA:The content of PKA of model group was greatly higher than blank control group and "Neiguan" group(P<0.05);The content of PKA of non-acupoint group was greatly higher than blank control group and "Neiguan" group(P<0.05).Conclusion1.Electro-acupuncture "Neiguan" can regulate the heart Rates of brady arrhythmia rats,so that abnormal cardiac rhythm(rate) tends to the normal.2.Electro-acupuncture "Neiguan" can releave the brady arrhythmia through changing the content of cAMP and the content of PKA.3.cAMP,PKA is related to the signal transduction,electro-acupuncture "Neiguan" can regulate the heart Rates through cAMP-PKA cell signal transduction.
