Objective:Observe electro-acupuncture’s effect on the insomnia rats’sleep, emotional activities and the neurotransmitter content of sympathetic adrenal quality system on HT7and point SP6, to reveal the psychological stress central mechanism of that how electro-acupuncture remedies insomnia.Methods:Divide the healthy clean grade SD rats to normal control group、model group and model electro-acupuncture group randomly. Make insomnia model by chronic restraint. Treat model electro-acupuncture group with electro-acupuncture on HT7and SP6. Observe and record every group rats’ ratio of0E%and0T%in the elevated plus-maze test, the stationary time in the rat tail suspension test and spontaneous activities of24hours. Detect every group rats’ blood plasma norepinephrine, epinephrine, dopamine level and norepinephrine, dopamine level in the hypothalamus and brainstem by ELISA test.Results:1. Changes in rats’emotion activities after treatment:①the stationary time in the rat tail suspension test has no difference between each group (P>0.05).②The ratio of0E%and0T%in the elevated plus-maze test,model group compared with normal control group the ratio both to be reduced (P<0.05),model electro-acupuncture group increased compared with model group (P<0.05), and to be no difference compared with normal control group (P>0.05)2. Spontaneous activity after treatment:①The daytime activity:model group compared with normal control group the daytime activity to be increased (P<0.05), model electro-acupuncture group reduced after treat by electro-acupuncture compared with model group (P<0.05), and to be no difference compared with normal control group (P>0.05).②The nighttime activity:model group reduced compared with normal control group (P<0.05) and model electro-acupuncture group increased compared with model group (P<0.05), no difference between electro-acupuncture group and normal control group (P>0.05)3. Norepinephrine and dopamine content in the hypothalamus and brainstem after treatment:①Norepinephrine content in the hypothalamus and brainstem of model group to be increased distinctly compared with normal control group (P<0.05);model electro-acupuncture group reduced obviously compared with model group (P<0.05), and have no difference compared with normal control group (P>0.05).②dopamine content in the hypothalamus and brainstem:compared with normal control group, model group is increased visibly, too(P<0.05);model electro-acupuncture group reduced obviously compared with model group (P<0.05), and have no difference compared with normal control group (P>0.05)4. norepinephrine, epinephrine, dopamine content in rats’plasma after treatment:①norepinephrine content:model group is increased signally compared with normal control group (P<0.05);model electro-acupuncture group reduced obviously compared with model group (P<0.05), and have no difference compared with normal control group (P>0.05).②epinephrine content:model group rats’ epinephrine content to be increased compared with normal control group (P<0.05);model electro-acupuncture group reduced obviously compared with model group (P<0.05), and to be no difference compared with normal control group (P>0.05)Conclusion:Chronic psychological stress insomnia rats’plasma norepinephrine, epinephrine, dopamine level and dopamine, norepinephrine content in the hypothalamus and brainstem increased, circadian activity to be abnormal, and in an anxiety and somnipathy state evidently. It can decrease the insomnia model rats’plasma norepinephrine, epinephrine, dopamine level and norepinephrine, dopamine content in the hypothalamus and brainstem by electric acupuncture on the points of HT7and SP6, to improve the anxiety and somnipathy state, and the content of norepinephrine, epinephrine and dopamine represents the excitability of the sympathetic adrenal medullary system, it reminds that the insomnia rats’excitability of the sympathetic adrenal medullary system to be increased and decreased after treating the insomnia rats by electric acupuncture on the points of HT7and SP6.
