OBJECTIVE:To develop a sensitive,accurate,simple HPLC method for the determination of common abuse drugs and their metabolites in unknown power,liquids and biological sample using solid-phase extraction by ultraviolet detector and provide the basis for the identification of the judiciary of Chongqing forensic examination institute.METHODS:The method was carried out on the Phenomenex Luna C18(4.6×250mm i.d,5μm).Two kinds of mobile phases were adopted in this study. Mobile phaseⅠwas consisted of acetonitrile and 30mmol·L-1 of NH4Ac buffer (10:90,v/v). Mobile phaseⅡwas consisted of acetonitrile and 30mmol·L-1 of NH4Ac buffer (55:45,v/v).The detective wavelength was set at 210nm, the flow rate was 1mL·min-1,the column temperature was room temperature and injection volume was 20μL.RESULTS:In plasma and urine sample, precision (RSD)varied from 1% to 6% and recoveris were between 98.1% and 101.0%.The standard curve and correlation coefficient of drugs in plasma are as follow, morphine Y=147.29X+5.2951, r=0.9999; MDA Y=21.202X+3.5002, r=0.9998;methamphetamine Y=29.653X+10.205, r=0.9997; MDMA Y=17.834X+5.5864, r=0.9975; 6-acetylmorphine Y=54.374X+12.791, r=0.9995;ketamine Y=264.52X - 1.3333,r=0.9998; The standard curve and correlation coefficient of drugs in urine are as follow, morphine Y=139.82X+8.058,r=0.9999;MDA Y=19.009X+7.8868,r=0.9999;methamphetamine Y=28.629X+10.857,r=0.9999; MDMA Y=16.12X+5.7768,r=0.9971; 6-acetylmorphine Y=52.918X + 8.632,r=0.9990 ;ketamine Y=236.81X + 6.4948,r=0.9998. The limit of detection (LOD,S/N=3)of morphine was 1.2ng, MDA 3.0ng, methamphetamine 2.0ng, MDMA 6.0ng, 6-acetylmorphine 2.4ng, ketamine 2.6ng.CONCLUSION:A RP-HPLC method was established ,which could separate and determine morphine, MDA, methamphetamine, MDMA,6-acetylmorphine and ketamine simultaneously. The method is simple, specific, reliable and suitable for the quantitative determination of these compounds in unknown powers, liquids and biological samples.
