Semen Momordicae, the dried and ripe seed of Momordica cochinchinesis Spreng (cucurbitaceae) is a poisonous Chinese medicine considered to exhitbit a cooling and resolvent effect, commonly used in remedy of wounds, bruises, swelling, hemonhoids and pus. Saponins are the major effective components in the medicine of M. cochinchinesis Spreng. In this thesis, the extraction and purification conditions were optimized, the antibacterial activity of saponins was studied, and the main component of saponins was analysed using High Performance Liquid Chromatorgraphy Electrospray/Mass Spectrometry (HPLC-MS). It will provide a theoretical basis for the application of saponins in M. cochinchinesis Spreng. The thesis is composed of four parts described as follows:In the first part, the chemical compositions in M. cochinchinesis Spreng, the pharmacological effects and the biological activity of the saponins, the ectraction, purification and analysis methods of the saponins, the contents as well as the sigificance of this thesis were introduced.In the second part, comparative study on the degrease effect of ultrasonic and flash-extraction; the methods of dipping extraction, ultrasonic and flash-extraction were used, and the extraction efficiencies of different methods were compared. The crude extract saponins were purified using macroporous resin and nbutanol alcohol extract, respectively. The results showed that ultraviolet method was of highest extraction efficiency, and flash-extraction consumed shortest time. The effects on the extraction efficiencies of cohol concentration, material/solvent ratio, and time were studied using orthogonal experiment design.The best extraction conditions were summarized as following: the alcohol concentration of 50%, the material/solvent ratio of 1:8, the extraction time of 40 sec.In the third part, the extraction, macroporous resin seperation, deposition, HPLC and TLC seperations, and vacuum freezing & drying technologies were utilized in extraction, purification and drying of the total saponins. The qualitative analysis results indicated that the above-mentioned extraction and purification methods were satisfactioy with the purity of 95.67 % of the purified saponin.In the partâ…£, comparative study on the in vitro antibacterial activity of raw medicinal material of M. cochinchinesis before and after pressing pulveratum process. The results indicated that the raw medicine material and pulveratum had strong inhibition effects on the growth of Candida Albicans, some inhibition effects on Staphylococcus aureus, no obvious inhibition effects on Escherichia coli and Pseudomonas aeruginosa. The pulveratum with the oil content of 10% and 20%, respectively, and saponins extracted from the seed of M. cochinchinesis had the stronggest inhibiting activity on bacterial growthes.In the partâ…¤, toxicity tests were carried out using different parts and the pulveratum of different oil content of the seed of M. cochinchinesis, The results indicated that the oil and seed capsule of M. cochinchinesis had no obvious toxicity; the toxicity of pulveratum was gradually decreased with the increase of its oil content.
