Caryopteris tangutica Maxim.belonged to Verbenaceae family distributes mainly in Shannxi, Sichuan and Gansu Province of China.It is a traditional Chinese medicine with the functions of activating blood,relieving pain,promoting digestion,regulating vital energy,removing toxic substances,dissipatating blood stasis and relieving itching.It's usually used to treat sores and eczema causing by hard-drinking.It also can regulate menstruation and cure leucorrhea,etc.In this paper we studied the antimicrobial and antioxidant activities of the extracts of C.tangutica,and analyzed the fat-soluble and volatile components of different organs of C.tangutica by gas chromatography-mass spectrometry(GC-MS).The main results were as follows:1.Volatile components from the stems,leaves and flowers of C.tangutica were obtained by hydrodistillation and Solid Phase Micro Extraction(SPME),and then analyzed by gas chromatography-mass spectrometry(GC-MS).Atotsl of seventy compounds were identified by hydrodistillation.Forty-two,forty-eight and thirteen compounds were identified from flowers,leaves and stems of C.tangutica,respectively.The main volatile compounds in flowers were trans-pinocarvyl acetate(46.69%),myrtenal(2.73%) and caryophyllene oxid(1.67%).The main volatile compounds in leaves were trans-pinocarvyl acetate(55.48%),beta.-cedrene(3.21%), caryophyllene oxid(2.73%).The main volatile compounds in stems were hexadecanoic acid (47.32%),trans-pinocarvyl acetate(7.47%) and caryophyllene oxid(0.96%).Forty-eight compounds were identified by Solid Phase Micro Extraction.Thirty-seven, thirty-four and tween-four compounds were identified from flowers,leaves and stems of C. tangutica,respectively.The main volatile compounds in flowers were trans-pinocarvyl acetate (63.85%),caryophyllene<(E)>(7.10%) and beta-himachalene(4.28%).The main volatile compounds in leaves of C.tangutica were trans-pinocarvyl acetate(76.11%),caryophyllene<(E)> (5.18%) and trans-chrysanthenyl acetate(2.53%).The main volatile compounds in stems of it were trans-pinocarvyl acetate(84.69%),caryophyllene<(E)>(3.20%) and trans-pinocarveol(2.72%).2.Fat-soluble constituents of the stems,leaves and flowers of C.tangutica were obtained by soxlet extraction,derivated by methyl esterification and identified by gas chromatography-mass spectrometry(GC-MS),fourty and thirty-one compounds were identified in flowers,leaves and stems,respectively.The main fat-soluble compounds in flowers were linolenic acid(27.37%), linoleic acid(14.35%) and trans-pinocarveol(8.40%).The main fat-soluble compounds in leaves were linolenic acid(25.48%),palmitic acid(12.77%) and linoleic acid(9.68%).The main fat-soluble compounds in stems were linoleic acid(9.8%),8,11,14-eicosatrienoic acid(8.05%) and palmitic acid (6.51%).3.In vitro antioxidant activities of the the extracts from different organs of C.tangutica were evaluated.Different samples(stems,leaves and flowers of C.tangutica) were extracted with ethyl acetate,methanol and water,respectively.The antioxidant activities were detected by four arrays (DPPH,β-carotene,OH·,O2- free radical-scavenging ability).The results showed that the methanol extract of the leaves exhibited the highest antioxidant activities among nine extracts of C.tangutica. It has higher contents of total phennolic compounds(2.08%) and flavone(1.80%) than others.The IC50 value of the methanol extracts from leaves of C.tangutica is 0.019±0.006(mg/mL),which is equivalent to synthetic antioxidant BHT.At the concentration of 0.12mg/ml,the ability of bleaching ofβ-carotene inhibitory activities reached 99%.The IC50 of OH·free radical-scavenging ability of total phennolic compounds from leaves of C.tangutica is 0.07±0.004(mg/mL),which is less than synthetic antioxidant Vc.The IC50 of O2.free radical-scavenging ability of flavone from leaves of C. tangutica is 0.03±0.006(mg/mL),which is higher than synthetic antioxidant Vc.4.The nine extracts of different organs(stems,leaves and flowers) of C.tangutica,were subjected to a screening for their possible antimicrobial activities using the method of Oxford cup. The volatile components of C.tangutica were subjected to screen for their possible antimicrobial activities using the method of 96-well microtitre plates.The results showed that all the extracts were found to inhibit the growth of tested bacteria.At the tested concentration,the antimicrobial activities of extracts from different organs of C.tangutica,to Mycobacterium smeganatis,are better than positive control tetracycline hydrochloride.The Candidatropicalis was sensitive to the volatile components of leaves of C.tangutica.The inhibition zone is 55.3±1.0 mm,MIC is 2.2 mg/mL,and MBC is 4.85 mg/mL,which is exceed to tetracycline hydrochloride(inhibition zone = 16.1±0.3 mm).
