Mosla chinensis Maxim cv. jiangxiangru is the aerial parts of plants belong to the member of Labiatae family, mainly distributed in Jiangxi province. Folk always used it to treat various cold, headache and other Summer diseases. Because of good efficacy, large-tonnage, it has become the mainstream products in Chinese mosla herb market. The earlier studies of Chinese mosla herb on chemical constituents and pharmacological activities are mainly concentrated in the volatile oil, other aspects of the research is less. In recent years, along with the advance of modern separation and analysis methods, the research on the chemical composition are turning to less volatile components such as polysaccharides, flavonoids, alkaloids, coumarin, and so on.Chinese medicine not only chemical composition is complex, and there are many different kinds of structure, but also contains several classes of different chemical properties of the material, the single detection method is difficult to realize effective analysis on its chemical composition, various analytical techniques combination is imperative. In order to deal with the complicated characteristics of traditional Chinese medicine chemical composition, at present has developed the HPLC-DAD/RI/FLR, GC-QQQ-MS, HPLC-ESI-QTOF/MS, HPLC-QQQ-MS and other rapid separation and analysis of research methods for the identification of the material. This issue chooses SPME-GC/MS and HPLC-ESI-QTOF/MS two coupled techniques optimize Mosla chinensis Maxim cv. jiangxiangru chemical composition analysis, main component, to provide certain theoretical basis for in-depth analysis of chemical composition analysis and clinical medicine safety and effectiveness. The concrete research content is as follows:Chapter 1: details about the research of chemical composition, separation, pharmacological effects of Mosla chinensis Maxim cv. jiangxiangru at home and abroad in recent years. of genera The chemical composition and pharmacological activity research is mostly in the volatile oil, the main volatile chemical composition is given priority to olefins and alcohols, the analysis method is always using GC-MS technology. The research of polysaccharide composition using gas chromatography before, because the sugar material molecular mass,less volatile, to analyze directly will be relatively difficult, and it also need to hydrolysis.Chapter 2: By using water bath circumfluence extraction jiang Chinese mosla herb polysaccharide aqueous solution, phenol-sulfuric acid method for the determination of total sugar content, total sugar content is 2.48%?2.45%?2.15%, respectively.Kjeldah method determination of protein content in MCJ water solution, its content is 8.48%?8.10%?9.01%. MCJ flavonoids with 60% ethanol solution under 75 ? temperature on the ultrasonic extraction, total content of flavonoids extracts determination by ultraviolet spectrophotometer, to 3.16%?3.22%?2.98%.Chapter 3: MCJ volatile constituents was extracted by the method of solid phase microextraction, the extraction was isolated and identified by the technique of combination of GC/MS. 73 kinds of volatile substances were preliminary identified, which alkenes compounds has 28 species accounted for 19.17% of total extraction of volatile compounds; alcohols have 10 species, but only 1.51% of total volatile components; phenolics are less in number, but the content was the highest, accounting for 67.84% of total volatile components, especially with thymol content was the high, accounting for 58.68% of total volatile components, is the main volatile constituents of MCJ. Test results are consistent with the reported results, but the method is simple and easy to operate, and has no need of reagent, the sample requirement is small, which can greatly reduce the time and steps needed to be processed before the experiment.Chapter 4: According to the report of the best extraction conditions of Mosla chinensis Maxim cv. Jiangxiangru polysaccharides extraction, physical and chemical properties were determined by chemical methods. The results show that PMJ is plant polysaccharide, the content of sugar is low. Crude polysaccharide total sugar content is only 15.87%, uronic acid is 11.16%, protein content is 1.51%. Using UPLC-ESI-QTOF/MS to analysis incomplete product of PMJ acid hydrolysis. With acetonitrile and 0.1% formic acid water as mobile phase, the positive and negative ions mode for UPLC-ESI-QTOF/MS and MS/MS analysis, it is concluded that the monosaccharide composition of PMJ is given priority to hexose, and it can detect nine Oligosaccharide from mass spectrometry. This method need not samples derived, can avoid the by-products from derivative interference, haing high sensitivity, good accuracy and less sample demand. It can be used as in-depth study and pharmacological action of polysaccharide in the late provide certain theoretical basis. Using DPPH, ABTS and FRAP to select effective components of antioxidant activity from five different solvent extracts. It is concluded that 70% ethanol extract objects have the highest antioxidant activity in vitro. Using UPLC-ESI-QTOF/MS detection of precise matching, quality measurement and possible formula, identified the main antioxidant activity substances of MCJ 70% ethanol extracts under positive and negative ions mode, a total of 54 substances were identified.
