Purpose:To further study the material foundation and mechanism of Astragalus mongholicus in immunological regulation by observing the effectiveness of Astragalus mongholicus and its various types of extractive on bone marrow depressed and injured mice resulting from chemotherapeutics—CTX.Material and method:1.Extract various composition of Astragalus mongholicus by use of polyamide and AB-8 macroporous resin.2.According to peripheral blood leucocyte count,divided 120 mice into 10 groups -uninfluenced,model,Astragalus mongholicus,extractⅠ,extractⅡ,extractⅢ,extractⅣ,extractⅤ,extractⅥand extractⅦgroups in randomication principle.Every groups has 12 mice with half of male.Uninfluenced and model group were given distilled water in the way of intragastric administration,the day before starting to make models,while other mice of medication administration groups were given correspongding medicine with the dosage of 0.15ml/kg in the same way,all for 14 days.From the day making models,all groups except uninfluenced one were given CTX in the way of intraperitoneal injection with the dosage of 150mg/kg,however,100mg/kg every other day,all for 6 times. Recorded the swimming time of every mice,2 hours after the last intragastric administration.24 hours later,killed all the mice for getting materials which were used to test peripheral hemogram,thymus and spleen index,IL-2 and TNF-αcontents and so on.Results:1.The peripheral blood cells count such as white blood cell,lymphocyte,red cell and hemoglobin of the myelosuppressive mice were degraded obviously.2.Symptoms of every treated groups resulting from CTX can relieve such as depression of spirit and decrease of activities,the descent of weight,the lack of shinning of fur.3.Compare with the model group,the swimming time of myelosuppressive mice in Astragalus mongholicus,extractⅠ,Ⅲ,ⅤandⅦgroups were obviously extended.4.Astragalus mongholicus,ExtractⅢcan increase the peripheral blood cells count of myelosuppressive mice.ExtractⅠ,Ⅱ,Ⅳ,Ⅵ,andⅦcan increase white blood cell count of mice,while ExtractⅤhas no character of effecting peripheral blood count.5.Compared with model group,the thymus and spleen index of mice in Astragalus mongholicus,ExtractⅠ,Ⅱ,Ⅲ,Ⅳ,ⅤandⅥhad increased,while ExtractⅦcan increase the spleen index of mice.6.IL-2 mice in Astragalus mongholicus,extractⅠ,Ⅱ,Ⅲ,Ⅳ,ⅤandⅦgroups increased,and TNF-αof mice in Astragalus mongholicus,extractⅠ,Ⅲ,Ⅵ, andⅦgroups increased too.Conclusion:1.Astragalus mongholicus and its extrative can protect the myelosuppressive mice in different degree.2.Astragalus mongholicus and its extrativeⅢare good at increasing the peripheral blood cells count;the extrativeⅠ,Ⅱ,Ⅳ,ⅥandⅦare expert in promoting white blood cell count in PB.3.Astragalus mongholicus and its extrative can promote the production of IL-2,TNF-α,improve the immunological regulation of myelosuppressive mice.
