K562/A02 cells were subcutaneously injected in the right side of the dorsal skin, and the K562 cells in the left side, the cells were inoculated for 1×107 cells of each injection. To observe the tumor and tumor growth characteristics. After successful establishment modle of the transplant tumor nude mice,divided those models into 5 groups:group of the physiological saline(group of K562), group of chinese herbal medicine big quantity, group of chinese herbal medicine Medium quantity, group of chinese herbal medicine small quantity, group of VER.To infuse mice stomach with YiQiYangYinJieDuFang and inject the belly cavity with ADM, examinate each tumor cell's store density of ADM and the expression of P-gp with FCM.Results:1) The rate of formation of K562 nude was 77.78%, the another was 73.33%, the tumors were formed in 7-13 days. There were no apparent difference between them; 2) The K562/A02 nude tumors cells and primary cells in vitro to ADM IC50 were 169.38ug/ml and 181.69ug/ml, and K562 IC50 were 3.47ug/ml and 3.61ug/ml, There were no apparent change in multiple drug resistance; 3) The expression of intracellular adria mycine were 50.24士3.51,36.95士3.20,19.97士0.47,when the dosage of YiQiYangYinJieDuFang were 0.8ml,0.4ml,0.2ml,the group of VER were 52.72士2.46;the group of physiological saline were 9.43士1.16;the group of K562 were 298.79士13.27. 4) the expression of P-gp were 226.16士25.97, 230.44士21.54,236.03士12.66, when the dosage of YiQiYangYinJieDuFang were 0.8ml,0.4ml,0.2ml, the expression of the group of VER were126.67士15.18,the expression of the group of physiological saline were 240.95士9.85.Conclusion:Firstly,The implant tumor, established by K562/A02 cells, remain holds the similar resistance activity to vitro. It could provide a reliable model for exploring the mechanism of multidrug resistance and reversling multidrug resistance.secondly,YiQiYangYinJieDuFang could induced apoptosis of the K562/A02 in nude mice and seemed that the effect was dependent on the dosage.YiQiYangYinJieDuFang could effectively reversed the resistance of transplantation tumor and increased the accumulance of intracellular adria mycine. Thirdly,the mechanism of reversal may be not related with inhibition of P-gp expression.
