| Objective To observe the retrograde amnesia in 8-arm radial maze in different injury times of cerebral concussion and to explore the change of pure cerebral concussion (PCC) and multiple cerebral concussion (MCC) on spatial cognitive behavior in rats. Methods A metallic pendulum striker device of closed brain injury was deployed to duplicate cerebral concussion and multiple cerebral concussion in Stragu-Dawley(S-D) rats. The animals were divided into pure cerebral concussion and 3MCC groups (n=8). One control group was used (n=8). Each group was assessed by food intake, entered the total number of arm with food, entered the food arm number, duplicate entry the number of arm without food, to explore the change of the reference memory and working memory. Result Compared with control, pure cerebral concussion and multiple cerebral concussion in food intake, entered the total number of arm with food, entered the food arm number, duplicate entry the number of arm without food are always declined, multiple cerebral concussion are more than pure cerebral concussion and recovery time more longer. Conclusion The cognitive- behavioral in rats was injured after cerebral concussion (CC). The damage of retrograde amnesia was worse in multiple cerebral concussion rats compared with pure cerebral concussion. Objective To establish the model of pure cerebral concussion(PCC) and multiple cerebral concussion(MCC) in rats, observe the changes of the expressing of ub post-PCC and post-MCC. To explore if MCC can increase the injury compared with PCC. Method The PCC or MCC model was duplicated by using a metallic pendulum -striker concussive device.96 female and male Sprague - Dawley rats were randomly divided into tweleve groups: PCCld,2d,4d,8d,16d,24d group and MCCld,2d,4d,8d,16d,24d group (n=6,in each group). One control was used (n=6). Using the immunostaining of streptavidin-peroxidase (S-P) method and image analysis system to detect optical density value of ub in the following areas, that is eg, HDB,MS,CPU, MHB, RSGb, S1BF, Aco,CA1-3, MHB,LDVL,VMH, IF,Pn and Ta. Result (1) The ub positive were located in most brain area.(2)The VOD of ub at median septum level in PCC:Loss of ub was significantly differently in the second day in the nucleus of cg,S1BF and the trough in forth day in the area of HDB,MS and in the 8th day in the area of CPU. (3)The VOD of ub at median septum level in MCC:Loss of ub was significantly differently in second day in the area of cg,S1BF,CPU,HDB. And the area of MS was significantly differently in 8th day. (4)The VOD of ub at hippocampus level in PCC:The trough of ub in the area of RSGb,S1BF,CA2,CA3,MHB,LDVL,VMH was in second day. CA1,Aco was significantly differently in 8th day.(5)The VOD of ub at hippocampus level in MCC:Loss of ub was significantly differently in second day in the area of RSGb,S1BF,VMH. The area of CA1,CA2,CA3,Aco was significantly differently in 4th day and in 8th day in the area of MHB,LDVL. (6)The VOD of ub at brainstem level in PCC:The trough of ub in the area of IF,Pn,Ta was in 8th day.(7)The VOD of ub at brainstem level in MCC:Loss of ub was significantly differently in second day in the area of IF,Pn. The area of Ta was significantly differently in the first day. Conclusion (1)The ub positive were located in most brain area. (2)After CC, Loss of ub was significantly differently in the area of cg,CPU,Aco,MHB,HDB,CA,LDVL,VMH.Loss of ub was significantly differently in second day and 8th day at median septum level and hippocampus level in PCC. The trough of ub at brainstem level in 8th day in PCC. Loss of ub was significantly differently in second day and 8th day at median septum level in MCC, the trough in second day and in forth day at hippocampus level and in second day at brainstem level in MCC. |
