PurposeTo explore the association of CETP gene TaqIB, D442G, I405V polymorphisms and the aldosterone synthase gene CYP11B2 gene-344C/T polymorphism with Essential Hypertension in Chinese Mongolian population.Method1) Sample selection of CETP gene polymorphism study:A total of 1927 Mongolian people, including 883(male 453,430 females) hypertensive patients and 1044 normal controls(male 372, female 672) from agriculture and pastoral areas in Tongliao city of Inner Mongolia, were included in this case-control study.2) CYP11B2 gene polymorphism in sample selection:A total of 2124 Mongolian people, including 925(male 460,465females) hypertensive patients and 1199 normal controls(male 421, female 778) from agriculture and pastoral areas in Tongliao city of Inner Mongolia, were included in this case-control study.3) SNP Genotyping:We identified the genotypes with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis and direct sequencing technology.Result1) The CETP gene polymorphism study showed the differences of TaqIB, D442G, 1405 V locus genotypes and alleles frequency distributions between Essential Hypertension and healthy controls were not statistically significant in men, women, and the total population.In the male group, the individuals carrying B2B2 genotype had higher high-density lipoprotein cholesterol level and lower total cholesterol, compared to those with B1B1 genotype (P=0.038, P=0.009);Individuals carrying B1B2, B2B2 genotypes had lower triglyceride levels compared to those with B1B1 genotype (P=0.014, P=0.016); individuals carried VV genotype had lower TC level, compared to those with II genotype (P=0.035).In the female group, individuals carried VV and IV genotypes had lower HDL-C level, compared to those with II genotype (P=0.003, P=0.011).The frequency of B2-G-I in the hypertensive group was significantly higher than in the control (P=0.005), so it is the susceptibility haplotype to EH.2) The CYP11B2 gene polymorphism study showed that-344C /T polymorphism was associated with essential hypertension in the Mogolian men, and CC genotype is more susceptibile to hypertension than TT genotype; In the male subgroup, individuals carried TC and CC genotypes had higher SBP than those with TT genotype(P=0.037, P= 0.007),and the DBP of those with CC genotype was significantly higher than that of people with TT genotype.Conclusion1) There are no correlations between CETP gene TaqIB, D442G,I405V polymorphisms single locus and essential hypertension in Chinese Monogolian population, but the hapolype B2-G-I is associated with essential hypertension, and it is susceptible to essential hypertension.2) CETP gene TaqIB, I405V polymorphisms are associated with lipid levels in Mongolian population.3) CYP11B2 gene-344C/T polymorphism is associated with essential hypertension in Chinese Monogolian men, CC genotype is susceptible to EH.
