Objective:Researching and evaluating the quality characteristic of different habitats, cultivated and uncultivated Salvia miltiorrhiza Bge. Analysing the distribution of parts of the effective components in Salvia's bark and in Salvia without bark. Studying the technology in the process of Salvia cutting. Researching the stability of effective water soluble components. We can find out the qulity influence factors of effective water soluble components, determine the rule of choosing and using Salvia, analyse the traditional quality judgement method, optimize the machining of cutting. We want to analyse the change law of water soluble components of Salvia miltiorrhiza Bge. and select evaluating targets so as to ensure Salvia miltiorrhiza Bge's quality.Methods:With HPLC measuring the content of the effective components, using the HPLC fingerprint characteristic to analyse, with statisticas analyzing the difference between different kinds or different environment. Using content measurement to analyse the distribution of parts of the effective components. Using orthogonal examination optimizing the machining process of cutting. Using acceleration test to analyse the change law of water soluble components of Salvia miltiorrhiza Bge.Results:1.Constant-temperature acceleration test:the content variations of the four water soluble components was studied on the fixed mesurement scheme. Results show that the degradation of the four water soluble components(Danshensu, Salvianolic acid B, Protocatechuic aldehyde and Caffeic acid) is the first order kinetic process. Salvianolic acid B can decompose into Danshensu. The result means that the main mesurement object of the water soluble components shoud be Salvianolic acid B and other effective water soluble components.2.With the mesurement of the effective components(Danshensu, Salvianolic acid B, Protocatechuic aldehyde, Caffeic acid, cryptotanshinone and Tanshinoneâ…¡A), we found out that there is no significant difference of the content of Salvianolic acid B between the samples in different producing area. But there is significant difference of content of the other three components between the samples in different producing area. There is significant difference between cultivated and uncultivated samples. The content of Salvia Danshensu, protocatechuic aldehyde, caffeic acid in the amount of wild species of Salvia is in higher levels but in cultivars is in lower levels; The content of Salvianolic acid B in the amount of wild species is in the lower levels but in cultivars is in higher levels.3.From the analysis of the HPLC fingerprint characteristic, we can find out that there are similar fingerprint peaks with pretty much the same peak shape and retention time between the wild and cultivated Salvia miltiorrhiza Bge, so can we find out the same conclusion between the samples in different producing area. Using the "chromatographic fingerprint evaluation system,2008 Edition" software, computing, similarity coefficients were over 0.97.4.The distribution of effective components between Salvia rough skin and the part without rough skin:The effective components were measured by HPLC in Salvia rough skin part, the part without the rough skin and the original medicinal part. The results showed that the distribution of the components is in different parts of the roots, which the fat-soluble components and Protocatechuic aldehyde, Caffeic acid mainly exist in the parts of rough skin, Danshensu and Salvianolic acid B mainly exist in the part without the rough skin.5.Orthogonal examination optimizing the machining process of cutting:The results showed that pressure and the water quantity are the factors of the content of the water-soluble components. The method of decompression can be used to reduced the loss of the content of the water-soluble components.6.Preliminary stability research:The stability of Salvia miltiorrhiza Bge was studied through accelerating test based on the fixed mesurement scheme of the four water soluble components. The mesurement results were compared with the original data. The content of Danshensu was obviously increased, while The content of Salvianolic acid B was obviously decreased. The results showed that the quantity of Danshensu isn't qualified with the quality of Salvia miltiorrhiza Bge.Conclusion:The HPLC measurement which can measure the contents of six kind effective components simultaneously proves to be speedy and exact. The fingerprint characteristic can include the mutual character of different kind, field and type. The difference of components between different kinds and different part can guide us utilizing the resources more reasonablely. We should pay attention to the quality of Salvia miltiorrhiza Bge. seed and its cultivating, the preparation technics of cutting, the expiry date and the storage condition. Definiting the evaluating targets and the study meets the anticipated objective.
