Objective:Research on the quantified fingerprint spectrum and efficacy of water-soluble components of Salvia Miltiorrhiza,establish the water-soluble components of Salvia Miltiorrhiza spectrum-effect related mathematical model,to explore the effective substance of water-soluble components and activity quantized evaluate system of Salvia Miltiorrhiza,in order to evaluate the quality more scientific,objective and comprehensive.Methods:Salvia Miltiorrhiza were collected from different areas in the country,build the quantified fingerprint spectrum of water-soluble components of Salvia Miltiorrhiza by optimize the preparation method of test solution and inspect the chromatographic condition such as wave length,column temperature,flow rate and interal standard,methodology examination,analyze the fingerprint spectrum by cluster analysis,principal component analysis and similarity analysis;analyze the components of fingerprint spectrum preliminarily by HPLC-MS;to establish the zebrafish intersegmental blood vessel?ISV?damage model by PTK787,establish the zebrafish heart injury model by terfenadine and establish the zebrafish caudal artery stenosis model by FeCl3,curing by different water-soluble components of Salvia Miltiorrhiza,determine the ISV,SV-BA,blood artery diameter and blood flow velocity,calculate the growth rate of ISV,heart repair,vasodilatation and rate of promote;analyze the sensitivity of the efficacy of water-soluble components of Salvia Miltiorrhiza by grey correlation degree analysis,demine the incidence of different pharmacodynamics indexes by comput the relevancy and relevant polarity.Establish the water-soluble components of Salvia Miltiorrhiza spectrum-effect related mathematical model by LS-SVM,built the quality evaluation system through blind authentication.Results:Confirm to prepare the test solution by water,establish the quantified fingerprint spectrum of water-soluble components of Salvia Miltiorrhiza by optimized condition,the RSD of precision test,stability test,repeatability test less than 3.0%;test 63 batches of Salvia Miltiorrhiza and select 9 common peaks;the range of similarity of different samples is 0.9281,the similarity range of different samples and control fingerprint is 0.9911,the samples are partitioned into 7 category by cluster analysis,determine 2 principal components,permutation and estimate the good and bad of different samples;the common peaks of fingerprint spectrum are identified by HPLC-MS,the number 1-9 peaks are Salvianic acid a sodium,Protocatechualdehyde,Salvianolic acid H,Rosmarinic acid,Alkannic acid,Salvianolic acid B,Salvianolic acid E,Isosalvianolic acid B/Isosalvianolic acid E,Salvianolic acid A;To establish the zebrafish intersegmental blood vessel?ISV?damage model by 0.3?g/mL PTK787,establish the zebrafish heart injury model by 2.5?g/mL Terfenadine and establish the zebrafish caudal artery stenosis model by 50?g/mL FeCl3,give different water-soluble components of Salvia Miltiorrhiza to observe the impaction,the range of the growth rate of ISV is 20%-85%,the range of the rate of heart repair is 29%-84%,the range of the rate of vasodilatation is 22%-75%,the range of the rate of promote is 7%-55%,compared with the normal group the treatment groups can promote the vascular development,protect heart and diastole the caudal artery obviously?p<0.05?;the 9 water-soluble components of Salvia Miltiorrhiza play a singnificant role in promoting the vascular development,protecting heart and diastoling the caudal artery by grey correlation degree analysis.Sort the contribution of ingredients to the efficacy of medicine by compute the comprehensive score.Built the spectrum-effect related mathematical model,the average relative prediction deviation is 5.04%.Conclusion:The method of Salvia Miltiorrhiza water-soluble components HPLC quantified fingerprint spectrum is simple,accurate,stable,the fingerprints has large amount of information and high accuracy,the pharmacodynamics test is accurate stable and sensitive,reveal material basis by identifying the common peaks and grey relational analysis,the spectrum-effect related quality evaluation system is feasible and stable which can provide a reference for the quality evaluation of Salvia Miltiorrhiza.
