1 Dioscorea zingiberensis C.H.Wright, also known as Huangjiang,is a unique plant growing in China. Huangjiangsu A is a single steroid compounds which was isolated from it. As there are many saponins whose structures are quite similar to Huangjiangsu A exist in the rhizome of Dioscorea zingiberensis C.H.Wright, the isolation and purification of the reference substance of Huangjiangsu A are very difficult. In this paper, we did a detailed research on the methods of extraction and separation of Huangjiangsu A. Three methods of its purification: the silica gel column chromatography, ODS silica gel column chromatography and preparative high performance liquid chromatography, were compared,and the qulity standard of Huangjiangsu A was drafted.2 Apocynum venetum belongs to Apocymaceae, Leaves of which are considered as a traditional, herbaL medicine.A capillary zone electrophoresis (CZE) method has been developed for the simultaneous determination of kaempferol, quercetin-3-O-β-D-glc(2â†'1)-β-D-glucose, kaempferol-3-O -β-D-glucose, isoquercitrin and quercetin in Apocynum Venetum leaves from different regions. The analytical conditions were optimized through investigation of the effects of buffer components, buffer pH and buffer concentration. These five flavonoids can be well-separated within 15 min at a working voltage of 20 kV and detection wavelength of 340 nm in a 50 mmol/L borate buffer (pH 9.3). The method is simple, rapid and sensitive with the small amount of sample and solvent required and ease of cleaning the column. Samples of Apocynum Venetum extracts were analyzed using the validated method,which is useful for its quality control. Through the determination by capillary zone electrophoresis (CZE), we found that quercetin-3-O-β-D-glucose and quercetin-3-O-β-D)-glc(2â†'1)-β-D-glucose are the main constituents of flavonoids in Apocynum venetum leaves, whose contents are up to 5.7‰and 3‰respectively. Flavones content is more and higher in Apocynum venetum leaves of Shan Xi province and Xin Jiang province than that of other habitats.
