The present study consists of two parts. The first part is about the expression and significance of ER,PR,CA215 in PTC. Objective:The aim of this study was to determine the expression of ER/PR and CA215 in human PTC and to explore the correlation of these proteins with the clinical features of PTC. Methods:Taking 69 samples of fresh PTC and 7 samples of nodular goiter from surgery, the study detected the expression of ER and PR in human PTC by immunohistochemistry and the expression of CA215 in 43 cases of these samples. The correlation of ER, PR, and CA215 expression with the biological characteristics of tumors was analyzed. Results: The ER and PR immunoreactivity were detected as 52.17% and 46.38% in PTC, respectively. There was no expression of ER and PR in nodular goiter and normal thyroid tissues around cancer. The expression of ER and PR was not associated with sex and tumor size. The expression of ER was negatively associated with age and the expression of PR was positively correlated with tumor stage, capsular invasion and lymph nodes metastasis (P<0.05). The rate of CA215 expression was 41.86% in PTC, which was positively related with the tumor size. Conclusion:These results indicated that estrogen might either stimulate the growth of cancer cells at the early stage of PTC, or stimulate initial production of PTC. Thus ER expression might be a sign of early stage and good prognosis in PTC. In addition, these results indicated that progesterone might inhibit tumor cell growth by PR in young patients, but PR expression in PTC might indicate that higher malignance and worse prognosis for elder patients. The study also found that the expression of CA215 occurred in nearly half of the PTC, which might provide a basis for CA215 antibody to be used as anticancer agents for refractory PTC.The second part is about the establishment of thyroid carcinoma cell line. Objective: The study was performed to establish PTC cell line in vitro, which could provide an experimental model for studying biological characteristics of PTC. Methods:The 9 cases with comparatively larger tumor sizes out of the 69 specimens were primary cultured in vitro. The expression of thyroglobulin (TG),cytokeratin (CK7) and ER by immunocytochemistry were detected. Results:There were 6 cells growing successfully in vitro for about 3 months. The six cell lines showed typical epithelial morphology. All the 6 cells expressed CK7 and some cell lines expressed TG and ER. Conclusion:It was easy for primary cultured PTC cells to grow and they grew well in the first few generations, but they grew limited in vitro. There were different ER, PR and CA215 expression in different cell lines, which represented different degrees of differentiation in tumor cells.
