Globus hystericus is the frequently-occurring disease of stomach intestine functional disease.It unconsciously presents as psychiatrical and neurological symptoms and globus sensation with patients'condition waving,lingering and difficult to cure.It has a bad influence on work and life of amplitude patients.At present,the study on its pathogenesis and ideal treatment is insufficient.Exterior and interior research indicated many severe emotional disorder to exist of the Globus hystericus patient,and account majority was anxiety-depression patient,so anti-depression play the crucial effective on to cure globus hystericus.Now Pinellia and Magnolia Bark Decoction has obtained a good effect on Globus hystericus and Depression.BanFo Nanometer pellets which is based on Pinellia and Magnolia Bark Decoction and finger citron and pseudostellariae product with the new nanotechnology. To research the dose–effect relationship of Pinellia and Magnolia Bark Decoction on anti-depressant effect .maked foundation for further study BanFo Nanometer pellets effect on Globus hystericus.For further study and trying to find out the toxicity of the BanFo Nanometer pellets,the study whcih is subsidized by Jiangsu Scinence And Technology Department.anti-depressant effect research【Aim】To observe the effect of Pinellia and Magnolia Bark Decoction on neuroendocrine and behavior of chronic stress depression rats. To research the dose– effect relationship of Pinellia and Magnolia Bark Decoction on anti-depressant effect .【Methods】seventy male SD rats were randomly divided into seven groups: normal control group,model group, Fluoxetine group and Pinellia and Magnolia Bark Decoction extreme low,low,mid and high groups. All rats except the control group were singly housed and exposed to an unpredicted sequence of mild stressor. The behavior of rats was detected by open - field test weight and sucrose preference test , and the levels of COR,ACTH in blood serum in different groups were observed by RIA.【Results】Compared with normal control group , the rats in the model group showed decreased body weight , lower sucrose water consumption test, reduced crossing and rearing scores significantly. Results also demonstrated that Fluoxetine, Pinellia and Magnolia Bark Decoction all groups can improve behavioral disorder. The concentration of blood serum COR,ACTH in stress -model rats compared to control groups haven't obviously diffierently. the increase of doses accompanied by gradually adding effect in the 0.45g/kg~2.3g/kg doses.【Conclusion】Chronic unpredicted mild stress can affect rats'behavior and cause depression. Pinellia and Magnolia Bark Decoction has antagonism against it and has some anti-depressant effect . the increase of doses accompanied by gradually adding effect in the 0.45g/kg~2.3g/kg doses.experiment on toxicity【Aim】To observe the acute toxicity and long-term toxicity of Banfo Nanometer pellets【Research contents】(1) experiment on acute toxicity of Banfo Nanometer pellets(2) experiment on long-term toxicity of Banfo Nanometer pellets【Methods and Results】(1) Experiment on acute toxicity40 rats was divided into experimental group and comparison group by chance,each group was 20,male and female was equal. Most dense Banfo Nanometer pellets solution (24g/kg) was gived to rats by gavage and observed weight,behaving and death of rats for two weeks.there is no obvious toxic symptom and death .observed continuously for a week,rats is alive, activity is freely,hair is slippery,bite and sup is normal,weight is increasing,nose,eyes and mouth do not have abnormal secretion.Anatomizing rats,observing main viscera by naked eyes and microscope,there is no obvious change in pathology. (2) Experiment on long-term toxicity80 mices was divided into high, mid,low dosage group and comparison group by chance,each group was 20. Banfo Nanometer pellets (10, 5.0, 2.5g/kg )was gived to high, mid,low dosage mices by gavage for 90 days,comparison group do not give medicine. Observing body's sign,behavior and activity,grand excrete,breath,dejection and appetite everyday,weigh mices once a week.24 hour after given medicine,take out 12 mices from each group,mices were anaesthesiaed by 2%Pentothal, blood index and blood biological indexes were inspected,mices were separated systemly,vicera was taken out and computed it's coefficient,organizations were inspected in pathology and histology.Leaving mices were executed after 2 weeks and taken those inspect,in order to know toxicity's reversible degree and lingering toxicity,Stat and analyse was taken by SPSS13.0,observe mainly the difference between medicine group and comparison group. There is no significant difference in food, behavior,weight,blood index and blood biological indexes,organ coefficient and organ pathology between the experimental and the control group.【Conclusion】Banfo Nanometer pellets solution(24g/kg) was gived to rats in experiment of acute toxicity,it is 240 times of adult does,there is no obvious acute toxic reaction. Banfo Nanometer pellets (10,5.0, 2.5g/kg)was gived to high, mid,low dosage mices ,it is 100, 50, 25 times of adult does.medicine was given according to set period,there is no obvious toxic reaction in growth, weight,blood index ,blood biological indexes,organ coefficient and organ pathology when stopping medicine and convalescene.As a result,there is no toxic reaction and lingering toxicity on rat and mice for Banfo Nanometer pellets.So it is safe at clinical dosage of Banfo Nanometer pellets.
