Purpose:Of this study was to explore the points of rabbit MSCs transplantation on ischemic myocardial cells ability to repair and reconstruction,and further research MSCs acupoint injection with peripheral venous injection of MSCs results compared,to explore a new and effective stem cell transplantation to repair damaged myocardium method.Material and method:With left ventricular branch ligation method of the establishment of acute myocardial infarction model, Japanese white rabbits what have been established myocardial infarction model were randomly divided into 5 groups. Model group, 0 days acupoint injection of stem cell group, 7 days, stem cells, acupoint injection group, 0 days of intravenous stem cell group, 7 days a group of intravenous injection of stem cells. The infarct zone Brdu immunohistochemistry observe the acupoint transplanted MSCwhether migrated to the infarct zone. Brdu staining of the cells stained positive for cardiac-specific troponin T staining in order to observe whether the differentiation of cell migration and differentiation of nature.And using Metamorph Ofline image analysis system, each slice randomly selected 5 high power field (40×), measured brdu positive cells in the Optical density Average.Results:The point of bone marrow-derived mesenchymal stem cells in the infarcted region can be seen staining the cell nuclei Brdu brownish yellow, indicating transplanted MSCs can migrate through the points to the infarct area. And these cells stained Brdu cardiac-specific troponin T staining (cTnT)-positive, indicating these cells have migrated to differentiate into cardiac cells.But control group, no Brdu-positive cells mark. In 0 day injection of bone marrow-derived mesenchymal stem cells, brdu-positive cells, the average optical density value, acupoint injection group was better than intravenous injection group. P<0.05 In 7 days injection of bone marrow-derived mesenchymal stem cells, brdu-positive cells, the average optical density value, acupoint injection group and the intravenous injection group are no significant difference.Conclusion:1. Point injection of bone marrow-derived mesenchymal stem cells can migrate to the infarct zone, and can differentiate into myocardial cells in the infarct.2. In 0 days injection of bone marrow-derived mesenchymal stem cells and it migrate to the infarct area, the memory of living in the infarct region to differentiate into cardiomyocyte-like cells, effect of acupoint injection is superior to intravenous injection.3. In 7 days injection of bone marrow-derived mesenchymal stem cells and it migrate to the infarct area, the memory of living in the infarct region to differentiate into cardiomyocyte-like cells, acupoint injection and intravenous injection showed no significant difference.
