On The 2nd. Lung Suppression Rather Experimental Studies Of Mice With Lewis Lung Cell Apoptosis

Objective:By studying the effect of the self-made Feiyining2 on Lewis lung cancer mice C57BL / 6 in the living conditions,the tumor inhibitory rate,the tumor cell morphological changes and the apoptotic gene p53, Bcl-2 expression to explore the mechanism of treatment of lung cancer by Yiqiyangyinjiedusanjie.Methods:To plant the Lewis lung cancer to 40 C57BL/6 mice by routine immunization, after five days, select the succeeded tumor-bearing mice to randomly divide into four groups: Model group, traditional Chinese medicine group, western medicine group and integrated group. Every group has 10 mouse. Take no planted cancer mice 10 as the blank group.After five days, to give each group medicine early morning when they are fasting. Gavage once daily for 12 days: traditional Chinese medicine group was given Feiyining 2, 16.5g/kg/d, 0.4ml. Other groups were given normal saline. Intraperitoneal injection by once every other day for 3 days: Western medicine group was treated with cisplatin injection by 0.13ml, other groups were given normal saline. Integrated group was given traditional Chinese medicine and cisplatin injection.Observe their daily life conditions and tumor growth, pathological changes of tumor tissue by HE staining, and electron microscopy of apoptosis, and the expression of apoptotic gene p53, Bcl-2 by Western Blot.Results:1. The prescription of Fei yining 2 can improve the general condition of Lewis lung cancer. Compared the model group, traditional Chinese medicine group and integrated group have more numbers of independent activities and standing by five minutes. The difference is statistically significant (P <0.05). But western medicine group reduce (P <0.05). Compared integrated group, traditional Chinese medicine group is no significant difference (P >0.05). 2. Experimental groups can inhibit tumor cell growth, tumor weight less than the model group. The difference is significant (P <0.05).The tumor inhibition rates: Integrated Group (50.37%), western medicine group (47.66%), traditional Chinese medicine group (35.05%).3. By the light microscope: Compared with model group, in the Chinese medicine group, western medicine group and integrated group, Tumor cell growth is limited, shaped tumor cell is low, can be seen"crescent"nuclear. By electron microscope, all experimental groups can be seen typical changes of apoptosis: apoptotic bodies.4. Western Blot assay results: Western group, traditional Chinese medicine group and integrated group can cut the mutant p53, Bcl-2 protein expression, Compared with model group, the differences are statistically significant (P <0.05).Conclusion:1. Fei yining 2 can improve the survival of tumor-bearing mice.2. Fei yining 2 has the role of treatment of lung cancer, and can directly inhibit the growth of Lewis lung tumor cells in mice.3. Combination with Cisplatin, Fei yining 2 can reduce the toxic side effects, improve their living conditions, play a synergistic role in attenuation.4. Fei yining 2 can induce lung cancer cells apoptosis.5. Fei yining 2 can down mtp53 Ning 2 and Bcl-2 protein expression. It may be one of the mechanisms of its anti-lung cancer.
